摘 要:目的:旨在通过动物实验观测参草通脉颗粒对慢性心衰大鼠的疗效。方法:选用健康雄性SD大鼠,采用冠状动脉结扎术配合饥饿、游泳等方法造成慢性心衰的动物模型。将造模成功的大鼠随机分为4组:模型组、中药大剂量组、中药中剂量组和西药组,每组8只;另设一组作为正常组。喂养8周后,观察大鼠一般情况、呼吸,心率的变化,测定心功能;用酶联免疫法(ELISA法)测定血清PRA,AngⅡ,血栓素B2 PAⅠ-Ⅰ;用透射电镜观察心肌细胞超微结构的变化。结果:中药大剂量组和中剂量组与西药组状态改善情况相似,P>0.05,无明显差异。中药大剂量组和中剂量组与西药组大白鼠用药后心率呼吸基本接近正常,组间比较P>0.05,无显著性差异。模型组大鼠心功能明显降低,与正常组比较,P<0.01;西药组和中药中剂量组疗效明显,与模型组比较,P<0.01,有显著差异;西药组和中药中剂量组之间比较,P>0.1,无显著性差异。而大剂量组疗效相对较差。PRA、AngⅡ均是中药组低于模型组(P<0.01);中药大剂量组与西药科素亚组比较,P>0.1;中药中剂量组低于西药科素亚组,P<0.01。血清血栓素TXB2、PAⅠ-Ⅰ模型组高于正常组,P<0.01,西药低于模型组,P<0.01,血清血栓素TXB2中药大剂量、中剂量组均低于西药,P<0.01;PAⅠ-Ⅰ中药中剂量组低于与西药组,P<0.01;大剂量组与西药组比较无差异。正常组显示心肌纤维排列整齐细胞数正常,染色比例正常,模型组显示心肌纤维紊乱,中药组和西药组显示细胞核基本正常。结论:参草通脉颗粒能改善心衰大鼠一般状态,降低血浆中PRA、AngⅡ、TXB2、PAⅠ-Ⅰ水平,改善大鼠心肌形态结构,以此来改善和纠正心衰。且以中剂量为好。Objective:To observation through animal experiments aimed at Shencao Tongmai Granules efficacy on rats with CHF. Methods:Male SD rats with coronary artery ligation and hunger,swimming to create an animal model of CHF. The successful model rats were randomly divided into four groups:model group,high dose group,middle dose group of Chinese medicine and western medicine group,each eight rats;separate one group as a control group. After 8 weeks of feeding,observerespiration,heart rate changes in rats,measuring heart function;serum PRA,AngⅡ,thromboxane B2 PAⅠ-Ⅰ enzyme-linked immunosorbent assay(ELISA);myocardial cells with ultra-TEM changes in microstructure. Results:high-dose group and middle dose group and western medicine group status to improve the situation is similar P〉0.05. No significant difference. The high-dose group and middle dose group and western medicine group about rats breathing and heart rate is close to the normal(P〉0.05),no significant differences. Model group significantly reduced heart function,compared with normal group P〈0.01;western medicine group and middle dose group in efficacy,compared with model group P〈0.01,significant difference;compare P between western and traditional Chinese medicine dose group P〉0.1,and no significant difference. The relatively poor efficacy of high-dose group. PRA,AngⅡ traditional Chinese medicine group were lower than the model group(P〈0.01),high-dose group and Cozaar group P〉0.1,the middle dose was lower than Cozaar group P〈0.01. Serum thromboxane TXB2,PAⅠ-Ⅰ:model group than in the normal group P〈0.01,higher than the model of western medicine group P〈0.01,serum thromboxane TXB2:traditional Chinese medicine group lower than western medicine P〈0.01,PAⅠ-Ⅰ:middle dose group lower than in traditional Chinese medicine and western medicine group P〈0.01,there were no differences.
