摘 要:通过使用改良的TCM199无血清培养系统,在96孔细胞培养板中,每孔放入一个腔前卵泡等条件下,探索添加物黄体生成素对小鼠腔前卵泡卵母细胞体外生长发育的影响,旨在摸索一套适合小鼠腔前卵泡体外生长发育的培养体系。结果表明:随着浓度的增加,卵泡和卵母细胞的直径都不同程度的有所增长,其中200mIU/mL浓度组在卵泡培养的同期时间,卵泡和卵母细胞的增长值最大,与其他各浓度组之间存在显著性差异(P<0.05)。卵泡培养到第2天时,对照组(0浓度组)卵泡存活率最高,为94%。但是培养到第6天时,200mIU/mL浓度组卵泡存活率最高,为62.5%。从卵泡成腔率来看,卵泡培养到第4天时,300mIU/mL浓度组卵泡成腔率最高,为24.4%。培养到第6天时,200mIU/mL浓度组卵泡成腔率最高,为35.4%。从卵泡发生生发泡破裂率和排出第一极体来看,300mIU/mL浓度组均略高于200mIU/mL浓度组。实验结果证明200mIU/mL浓度组与其他浓度组相比卵泡存活率最高,成腔率最高。且卵泡及卵母细胞增长最快,但发生生发泡破裂率、排出第一极体率次于300mIU/mL浓度组。Based on the modified TCM199 serum-free culture system,with one preantral follicle in each hole of the 96-hole cell culture plate,the influence of additive luteinizing hormone(LH) on in vitro development of preantral follicles and oocytes of the mouse was researched in order to design a suitable culture system for in vitro development of preantral follicles. The results showed that with the increase of concentration,the diameters of follicles and oocytes grew and the growth of follicles and oocytes at the same period of culture was the largest at the concentration of 200 mlU/mL,which were significantly different from the growth at different concentrations(P〈0.05). On the second day of culture,survival rate of follicles at the concentration of 0 mIU/mL was the highest(94%) but the highest(62.5 %) at the concentration of 200 mIU/mL on the sixth day. Follicular cavity rate at the concentration of 300 mlU/mL were the highest(24.4 %) on the fourth day but was the highest(35.4%) at the concentration of 200 mIU/mI, on the sixth day of culture. Breaking rate of follicular germinal vesicles and discharge rate of the first polar body at the concentration of 300 mlU/mL were both slightly higher than those at the concentration of 200 mIU/mL. In conclusion,survival rate of follicles and follicular cavity rate at the concentration of 200 mlU/mL were much higher than those at other concentrations,and the growth of follicles and ooeytes was maximum but breaking rate of follicular germinal vesicles and discharge rate of the first polar body were only lower than those at the concentration of 300 mIU/mL.
