摘 要:目的探讨中药溪黄草水提取物对醋氨酚所致小鼠急性肝损伤的保护作用。方法 NIH小鼠随机分为6组,其中溪黄草(低、中、高剂量)/醋氨酚组分别灌服溪黄草水提物200、400、800 mg·kg-1,溪黄草对照组灌服溪黄草提取物400 mg·kg-1,正常对照组和醋氨酚模型组灌服相同体积的生理盐水,每次间隔12h,连续7d。末次给药后1h,醋氨酚造模组和溪黄草/醋氨酚组腹腔注射给予醋氨酚(350 mg·kg-1)造急性肝损伤小鼠模型,正常对照组、溪黄草对照组则腹腔注射相同体积的生理盐水,36h后处死小鼠,收集血液和肝脏组织,检测血清ALT、AST活性和SOD、H2O2、MDA水平。结果实验造模成功,与造模组比较,灌胃给予高、中、低不同剂量的溪黄草水提物,均能显著降低小鼠血清ALT、AST活性,升高肝组织中SOD水平,清除肝脏匀浆H2O2和降低肝脏MDA含量,且该水提物对不同指标的影响均呈现明显的量效关系。结论溪黄草水提物对醋氨酚所致的小鼠急性肝损伤具有保护作用,抗氧化作用可能是其保护作用的机制之一。Objective To investigate the protective effect of the aqueous extract of Rabdosia serra on acute liver injuryinduced by paracetamol in mice. Methods NIH mice were randomly divided into 6 groups,and the aqueous extract ofRabdosia serra(200, 400 and 800 mg·kg^-1per day) by gavage for 7 consecutive days was administered in the Rabdosia serra/paracetamol group. The aqueous extract with a dosage of 400 mg·kg^-1per day was given to mice in the Rabdosia serra controlgroup, and at the same time. physiological saline was administered in control group and the paracetamol group. One hour afterthe last dose of Rabdosia serra extract, a single dose of 350 mg·kg^-1paracetamol was given by intraperitoneal injection in theparacetamol group and the Rabdosia serra/paracetamol group, and the same dose of physiological saline was given in thecontrol group and the Rabdosia serra group. All mice were killed at 36 h after acetaminophen treatment. Serum samples andliver tissues were collected for biochemical analysis. Results The model was established successfully. Compared with thosein mice treated paracetamol alone, the aqueous extract of Rabdosia serra with different dosages could significantly decrease theALT and AST activities, increase hepatic SOD levels, and reduce liver H2O2 and MDA levels. And it showed a significant dose-dependent effect. Conclusions The aqueous extract of Rabdosia serra exerted a strong protection against paracetamol-induced liver injury in mice, the antioxidative effect should be one of its major protective mechanism.
