CIA小鼠血清中IL-21、IL-22、IL-23的变化研究

摘　　要：目的观察鸡Ⅱ型胶原诱导型关节炎(CIA)小鼠血清中IL-21、IL-22、IL-23的动态变化。方法 36只DBA1/J小鼠随机分为正常组12只(每时间点各6只)和模型组24只(每时间点各12只)。分别在CIA小鼠加强免疫7d及35d无菌摘眼球取血,常规分离血清,应用流式细胞术检测各组小鼠在加强免疫7d及35d血清中细胞因子IL-21、IL-22、IL-23的变化情况。结果模型组IL-21及IL-23在加强免疫7d显著升高,与对照组比较,差异有统计学意义(P<0.05;P<0.01),加强免疫35d与正常组比较差异无统计学意义(P>0.05);IL-22在病程进展中,模型组与正常组相比差异无统计学意义(P>0.05)。结论细胞因子IL-21和IL-23参与了CIA的发病,可能与类风湿关节炎的炎症进展有一定关系。Objective To observe the dynamic changes of levels of IL-21,IL-22 and IL-23 in serum of chicken type Ⅱcollagen-induced arthritis（CIA） mice.Methods The 36 DBA1/J mice were randomly divided into normal group（n = 6） andmodel group（n = 12）, the sera were taken from the eyeball blood and separating under sterile condition on the 7d and 35 d afterbooster immunization.The levels of cytokines IL-21,IL-22 and IL-23 were detected by flow cytometry. Results The levels ofIL-21 and IL-23 were significantly higher in the model group than that in the normal group（P〈0.05,P〈0.01）on the 7d afterbooster immunization, but there were no significant difference between normal group and model group on the 35 d after boosterimmunization（P 〉 0.05）. During the course of disease, the increase of level of IL- 22 in model group was not significantcompared to that of the normal group（P 〉0.05）.Conclusion IL-21,IL-22 and IL-23 paticipate in the pathogenesis of CIAand their variations at different stages of the disease are associated with RA.
