摘 要:目的研究苦参碱对人乳腺癌(michigan cancer foundation-7,MCF-7)细胞的促凋亡作用及对线粒体跨膜电位的影响。方法体外培养人乳腺癌MCF-7细胞,观察组细胞中加入不同浓度的苦参碱溶液,对照组细胞中加入等量的培养液。MCF-7细胞的凋亡与细胞线粒体跨膜电位采用流式细胞仪检测,MCF-7细胞的抑制率采用MTT的方法检测。结果苦参碱处理24h后观察组的MCF-7细胞凋亡率明显高于对照组,差异有统计学意义(P<0.05),且随着浓度的升高而升高;苦参碱对于MGF-7细胞抑制率的结果显示,随着时间与浓度的增加苦参碱对于MGF-7细胞抑制率效应也随之增强,且差异有统计学意义(P<0.05);苦参碱处理24 h后的罗丹明染色阳性数低于对照组,差异有统计学意义(P<0.05),表明线粒体跨膜电位与浓度呈负相关。结论苦参碱对人乳腺癌MCF-7细胞有促凋亡的作用,其促凋亡的作用可能是通过降低线粒体跨膜电位而使MMP的通道打开。Objective To study the effect and the analysis of matrine for induced apoptosis in breast cancer MCF-7 cells and mitochondrial transmembrane potential. Methods Breast cancer MCF-7 cells were cultured in vitro, then they were divided according to the random number to observation goup and control group. The observation goup were treated by different concentrations of matrine solution, and control group were added an equal amount of broth. Apoptosis and mitochondrial membrane potential of MCF-7 cells were detected by flow cytometry (FCM) detection, the inhibition rate of MCF-7 cells was detected by MTI'. Results MCF-7 cell apoptosis rate in observation goup was significantly higher than that in control group after 24h, and there were statistically significant ( P 〈 0. 05 ), it increased as the concentration had increased ; inhibition rate of matrine for MGFo7 cell showed that inhibition rate had increased with time and concentration of matrine, and there was a statistically significant ( P 〈 0. 05 ) ; Rhodamine staining positive number in observation group was less than the control group, and there were statistically significant ( P 〈 0. 05 ), and the number of positive Rhodamine staining increased as the concentration had increased. Conclusion Matrine can promote apoptosis for human breast cancer MCF-7 cells, the effect may be caused by reducing mitochondrial transmembrane potential and leaving the MMP channel open.
