摘 要:目的:观察中药升降散对脓毒症小鼠CD4+T辅助细胞Th1/Th2和Th17/Treg失衡的干预作用。方法80只小鼠随机分为对照组、假手术组、脓毒症组和升降散组,脓毒症造模后比较升降散干预前后不同时间段各组小鼠Th1、Th2、Th17和Treg细胞占辅助T细胞比例及Th1/Th2、Th17/Treg比值的变化。结果①Th1细胞比例:脓毒症组24 h内持续上升,升降散组24 h内逐渐下降,12 h和24 h脓毒症组均高于对照组、假手术组和升降散组( P<0.05或<0.01)。Th2细胞比例各时间段在各组之间差异均无统计学意义( P>0.05)。 Th17细胞比例:升降散组24 h内呈逐渐升高趋势;6 h脓毒症组显著高于对照组、假手术组和升降散组(P<0.01),12 h和24 h脓毒症组均高于对照组和假手术组(P<0.05或<0.01),12 h和24 h升降散组显著高于对照组和假手术组( P<0.01)。 Treg细胞比例:脓毒症组、升降散组24 h内呈逐渐升高趋势;各时间段假手术组、脓毒症组和升降散组均高于对照组(P<0.05或<0.01),24 h脓毒症组高于假手术组和升降散组(P<0.05或<0.01)。②Th1/Th2比值:各时间段脓毒症组均高于对照组(P<0.05或<0.01),升降散组与对照组比较差异无统计学意义(P>0.05),24 h脓毒症组高于假手术组和升降散组(P<0.05);Th17/Treg比值:各时间段脓毒症组均高于对照组和假手术组(P<0.05或<0.01),6 h脓毒症组显著高于升降散组(P<0.01),12 h和24 h升降散组显著高于假手术组和对照组(P<0.01)。结论升降散具有调节脓毒症小鼠CD4+T辅助细胞Th1/Th2和Th17/Treg失衡的作用。Objective To investigate the intervention of Chinese herb Shengjiang San ( SJS) for the imbalance of Th1/Th2 and Th17/Treg cells ratio in sepsis mice.Methods Eighty experimental mice were randomly divided into four groups: control group of 8 cases, sham operation group of 24 cases, sepsis group of 24 cases and TCM participation group ( SJS group) of 24 cases.After CLP of sepsis model, Th1, Th2, Th17 and Treg cells in the proportion of hole CD4 +Th cells and Th1/Th2, Th17/Treg ratio were compared respectively in each group before and after (6 h, 12 h and 24 h) SJS treatment.Results ①Proportion of Th1 cells was increasing in sepsis group and decreasing in SJS group at 24 hours.In sepsis group, it was higher than that in control group, sham operation group and SJS group at 12 hours and 24 hours (P〈0.05 or 〈0.01).There was no significant difference in proportion of Th2 cells among all groups at each time point(P〉0.05).Proportion of Th17 cells was increasing in sepsis group in 24 hours and it was higher in sepsis group than that of in control group, sham operation group and SJS group at 12 hours and 24 hours (P〈0.01).In SJS group, it was higher than in control group, sham operation group at 12 hours and 24 hours (P〈0.01).Proportion of Treg cells was increasing both in sepsis group and SJS group at 24 hours.In control group, it was lower than that in sham operation group, sepsis group and SJS group at each time point(P 〉0.05).In sepsis group, it was higher than that in sham operation group and SJS group at 24 hours ( P 〈0.05 or 〈0.01).②Th1/Th2 cells ratio in sepsis group was higher than that in control group, sham operation group and SJS group (P〈0.05 or 〈0.01) while there was no significant difference of Th1/Th2 cells ratio between SJS group and control group(P〉0.05)at each time point.In sepsis group, it was higher than in sham operation group and SJS group at 24 hours (P〈0.05).Th17/Treg cells ratio in sepsis group was higher than that in
