摘  要:目的为研究溴氰菊酯(deltamethrin,DM)对成年大鼠睾丸组织的毒性作用。方法将60只成年大鼠随机分为5组,即对照(玉米油)组和2.1、4.375、8.75、17.5 mg/kg DM染毒组,每组12只,连续灌胃染毒15 d。测定大鼠每日生精量和雄激素受体(AR)磷酸化蛋白水平,观察睾丸形态的变化。结果与对照组比较,8.75、17.5 mg/kg溴氰菊酯染毒组大鼠睾丸每日生精量均下降;各剂量溴氰菊酯染毒组大鼠睾丸组织中磷酸化AR蛋白表达水平和萎缩生精小管数均较高,生精小管数均减少,生精小管周长均缩短,差异有统计学意义(P<0.05)。且随着溴氰菊酯染毒剂量的升高,大鼠睾丸每日生精量、生精小管数和生精小管周长呈逐渐下降的趋势,睾丸组织中磷酸化AR蛋白表达水平和萎缩生精小管数均呈上升趋势。结论 DM可通过提高AR磷酸化水平来抑制AR的活性,导致大鼠睾丸组织损伤及生精能力下降。Objective To investigate the toxicity of dehamethrin (DM) to testis of adult male rats. Methods A total of 60 adult male Sprague-Dawley rats were randomly divided into five groups, 12 in each, treated with DM through gavage at the doses of 0,2.1,4.375,8.75 and 17.5 mg/kg respectively, once a day for 15 consecutive days. After 15-day treatment, the daily sperm production,morphologic changes of testis and phosphorylation of androgen receptor (AR) were studied. Results The rats treated with DM at 8.75 and 17.5 mg/kg body weight exhibited a significant reduction in testicular daily sperm production compared with the control group (P〈0.05). In DM-treated groups,the number and the perimeter of seminiferous tubules exhibited reduction, while the number of tubules exhibiting degeneration and phosphorylation of AR significantly increased, with statistical differences as compared with the control group (P〈0.05). Along with the increase of DM doses, testicular daily sperm production appeared a decreasing trend, as well as the number and the perimeter of seminiferous tubules, while phosphorylation of AR and the number of degenerated tubules increased. Conclusion DM can inhibit the activity of AR by increasing the phosphorylation of AR,leading to impairments of the structure of seminiferous tubules and spermatogenesis in adult male rats.
