摘  要:目的研究谷糠内壳结合态多酚(BPIS)的抗肿瘤活性。方法谷糠内外壳粉碎后,分别采用80%丙酮提取自由态多酚;残渣加入氢氧化钠消化后,用乙酸乙酯去脂提取结合态多酚。用福林酚比色法测定其成分含量,对谷糠内外壳不同形式多酚进行含量比较;MTT法评价谷糠多酚提取物对多种肿瘤细胞的体外抑制作用;用克隆形成实验评估了BPIS对肿瘤细胞存活的影响和对细胞群体形成集落或克隆的能力;使用荧光素标记的膜联蛋白(FITC-Annexin V)联合流式细胞术,检测BPIS对人肝癌细胞(Hep G2)、人宫颈癌细胞(He La)凋亡发生率的影响。结果谷糠内壳的结合态多酚(BPIS)与其他三种形式的多酚相比能够显著抑制Hep G2、He La、人乳腺癌细胞(MCF-7)、人结肠癌细胞(HCT-116)细胞的增殖;BPIS长期处理可显著减少癌细胞的克隆存活;随着BPIS浓度的增加细胞凋亡率增加。此外,BPIS对Hep G2和He La细胞的抑制作用与其浓度和作用时间呈正相关。结论谷糠内壳中的结合态多酚对多种癌细胞具有杀伤作用,它是通过诱导肿瘤细胞凋亡而抑制细胞的增殖。Objective To investigate the anti-tumor activity of bound polyphenols in inner shell(BPIS) from foxtail millet bran. Methods The soluble free polyphenols of external and inner shells from foxtail millet bran were extracted using 80% acetone. The bound polyphenols were from the residue after the extraction of free polyphenols. The residue was digested with sodium hydroxide. The remaining mixture was then extracted with ethyl acetate. Polyphenol contents of the bound and free polyphenols from external and inner shells were compared by the Folin-Ciocalteu assay. To evaluate the long-term effects of BPIS on cell survival and the ability to form colonies, the clonogenic survival assay was carried out. The effects of BPIS on the apoptosis of Hep G2 and He La cells were determined by Annexin V/PI double staining. Results The study showed that of BPIS from foxtail millet bran had anti-proliferative activities against Hep G2 cells, He La cells, MCF-7 cells and HCT-116 cells. It was indicated that the clonogenic survival of tumor cells was reduced after long-term exposure to BPIS, and there was a positive correlation between BPIS concentration and cell apoptosis. In addition, BPIS acted on Hep G2 and He La cells in dose-dependent and time-dependent manners. Conclusion The results demonstrated a broad-spectrum anti-proliferative property of BPIS. The mechanism may be associated with induction of apoptosis.
