摘 要:目的:建立柠檬苦素在大鼠肝灌流液中含量的测定方法。方法:SD大鼠行灌流手术,采用浓度为28.0μg·m L-1的柠檬苦素溶液对大鼠离体肝进行灌流,HPLC法测定灌流液中柠檬苦素的含量。结果:柠檬苦素在0.060~96.0μg·m L-1(n=9)范围内线性关系良好(r=0.998 8);定量限为50 ng·m L-1,方法回收率为93.6%~102.1%。结论:本方法快速、准确、简便,能够满足柠檬苦素在大鼠肝脏中药动学的研究。Objective : To develop a method for the determination of limoniin in the perfusate from isolatedrat tion nln. liver. Methods: The perfusion experiments were performed in SD rats, in which 28.0 μg·m-1, was infused into tile isolated liver. The resultant perfusate samples were assayed by HPI,C for determining Results: The calibration curves of limonin were linear in the range of 0. 060 ~ 96.0 μg· mL-1 (n = 9 0.9988). The limit of quantitation was 50 ng·mL-1. The relative recoveries were 93.6% ~ 102. 1%. Conclu-sion: The method is simple, sensitive and suitable for the determination of limonin content in the perfusate from i- solated rat liver.
