摘 要:目的 研究氯化钆(GdCl3)在脓毒症诱导大鼠ALI中对肺组织IL-10、巨噬细胞炎性蛋白2(MIP-2)表达的影响,探讨GdCl3对肺损伤的保护机制.方法 36只Wistar雄性大鼠随机平均分为3组:假手术组、脓毒症组和GdCl3治疗组.脓毒症组和GdCl3治疗组行盲肠结扎穿孔术构建脓毒症ALI模型.假手术组只翻动盲肠不结扎穿孔,GdCl3治疗组术后即刻经尾静脉注射2% GdCl3溶液(10 mg/kg).术后6h处死动物取样,ELISA法测定血清中IL-10、MIP-2水平,Western blot法测定肺组织IL-10、MIP-2的表达水平,观察肺组织病理结构变化,用SPSS 19.0统计软件统计结果.结果 与假手术组相比,脓毒症组、GdCl3治疗组肺组织内MIP-2表达水平明显升高(P<0.05);与脓毒症组相比,GdCl3治疗组肺组织内MIP-2表达水平降低(P<0.05);与脓毒症组相比,假手术组肺内IL-10表达差异无统计学意义(P>0.05),GdCl3治疗组IL-10表达升高(P<0.05);肺组织病理结果提示脓毒症组和GdCl3治疗组肺内大量炎性细胞浸润、肺间质水肿、肺内出血,但GdCl3治疗组症状较脓毒症组明显减轻.结论 脓毒症ALI时,肺组织MIP-2表达明显升高,大量炎性细胞浸润.GdCl3通过调控肺泡巨噬细胞使其促炎细胞因子MIP-2表达减少和抗炎细胞因子IL-10表达增加,减少中性粒细胞浸润,起到肺保护作用.Objective To study the influence of gadolinium chloride (GdCl3) in the expressions of interleukin-10 (IL-10) and macrophage inflammatory protein-2 (MIP-2) in lung tissue of rats with acute lung injury induced by sepsis,and to explore protection mechanism of GdCl3 on lung injury.Methods 36 male Wistar rats were randomly divided into three groups:sham-operated group,sepsis group,and GdCl3 group.Sepsis group and GdCl3 group underwent cecal ligation and puncture to build acute lung injury model induced by sepsis.Sham-operated group only fliped but not ligate and puncture cecal.GdCl3 group was immediately injected with 2% GdCl3 solution (10 mg/kg) through the tail vein after operation.The animals were sacrificed after six hours and sampled.The expressions of serum IL-10 and MIP-2 were assayed by the method of ELISA,the expressions of IL-10 and MIP-2 in lung tissue were measured by Western blot.The structural changes in lung tissue was observed.The statistical results were calculated with SPSS 19.0 statistical software.Results Compared with sham-operated group,expression of MIP-2 in lung tissues of sepsis group and GdCl3 group was significantly increased (P 〈0.05).Compared with sepsis group,expression of MIP-2 in lung tissues of GdCl3 group was lower (P 〈0.05).Compared with sepsis group,IL-10 expression in lung tissues of sham-operated group was no significantly different (P 〉0.05),but it was significantly increased in GdCl3 group (P 〈0.05).The lung pathology results showed that the inflammatory cell infiltration,interstitial pulmonary edema,pulmonary hemorrhage appeared in lung of sepsis group and GdCl3 group,but symptoms of GdCl3 group was significantly reduced compared with that of sepsis group.Conclusions MIP-2 expression is significantly increased and inflammatory cell infiltration appears in lung tissue in acute lung injury induced by sepsis.GdCl3 make expression of proinflammatory cytokine MIP-2 decreased and expression of anti-inflammatory cytokine IL-10 increased throu
