摘  要:目的建立HPLC法同时测定安神解虑颗粒中紫丁香苷、栀子苷、刺五加苷E、迷迭香酸4种有效成分。方法安神解虑颗粒75%甲醇提取液的分析采用Welch XtimateTMC18色谱柱(4.6 mm×250 mm,5μm);以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱;体积流量1.0 m L/min;柱温30℃;检测波长分别为265 nm(0~25 min),238 nm(25~35 min),220 nm(35~58 min)和330 nm(58~88 min)。结果紫丁香苷、栀子苷、刺五加苷E、迷迭香酸的进样量分别在0.022 8~0.285μg(r=0.999 5)、0.564~7.050μg(r=0.999 7)、0.074~0.925μg(r=0.999 1)、0.053~0.663μg(r=0.999 2)范围内线性关系良好,平均加样回收率分别为97.6%、97.4%、97.9%、97.3%,RSD分别为0.7%、0.8%、0.9%、1.1%。结论本方法简便、准确、重复性好,可为安神解虑颗粒的质量标准研究提供参考。AIM To establish an HPLC method for simultaneously determining four constituents,including syringoside,geniposide,eleutheroside E and rosmarinic acid,in Anshen Jielü Granules. METHODS The analysis was carried out on Welch xtimateTMC18column(4. 6 mm × 250 mm,5 μm). The mobile phase consisted of acetonitrile-0. 1% phosphoric acid solution in step gradient elution mode. The flow rate was 1. 0 m L/min,the column temperature was 30 ℃ and the detection wavelengths were set at 220 nm,238 nm,265 nm,and 330 nm,respectively. RESULTS The experimental data showed a good linearity in the range of 0. 022 8- 0. 285 μg(r =0. 999 5) for syringoside,0. 564- 7. 050 μg(r = 0. 999 7) for geniposide,0. 074- 0. 925 μg(r = 0. 999 1)for eleutheroside E,0. 053- 0. 663 μg(r = 0. 999 2) for rosmarinic acid,respectively. The average recoveries were 97. 6%, 97. 4%, 97. 9%, and 97. 3%, respectively. The RSDs were 0. 7%, 0. 8%, 0. 9%, and1. 1%,respectively. CONCLUSION This method with strong specificity is simple,accurate and reproducible,and it can provide the reference for quality standard for Anshen Jielü Granules.
