摘 要:目的比较研究不同产地海螵蛸的指纹图谱,并测定其中核苷类成分的含有量。方法海螵蛸Na Cl水溶液采用高效液相色谱法,色谱柱为资生堂PAK-C18(4.6 mm×250 mm,5μm),流动相为0.01 mol/L磷酸二氢钾水溶液,等度洗脱,检测波长254 nm,柱温25℃。用相似度评价系统软件计算得到各批药材的相似度。结果海螵蛸中的尿嘧啶、次黄嘌呤和黄嘌呤分离度良好,线性范围分别为1.25~12.50μg/m L、1.28~12.80μg/m L和5.05~50.50μg/m L。25批样品(来自浙江、辽宁、云南、山东、广西、福建)相似度均大于0.9;广东的样品相似度在0.97~0.78,差异较大;海南的样品相似度均为0.88左右。结论本研究建立的HPLC指纹图谱方法可有效地区分海螵蛸及其他贝壳类海洋药材。AIM To compare the HPLC fingerprints of cuttlebone(Sepiae endoconcha) from different regions and determine the nucleotides content. METHODS HPLC analysis of Na Cl solution of cuttlebone was carried out on Shiseido PAK-C18(4. 6 mm × 250 mm,5 μm) with a mobile phase of 0. 01 mol/L potassium dihydrogen phosphate solution in an isocraic elution manner,the flow rate was 0. 65 m L/min,the sample amount was 20 μL,the detection wavelength was set at 254 nm and the column temperature was maintained at 25 ℃. The similarity software was used to evaluate the fingerprint similarity among the batches. RESULTS The method had good linear relationship within the range 1. 25- 12. 50 μg/m L for uracil,1. 28- 12. 80 μg/m L for hypoxanthineand,and 5. 05-50. 50 μg/m L for xanthine. The similarity of twenty-five batches was greater than 0. 9 collected from Zhejiang,Liaoning,Yunnan,Fujian Province,and Guangxi region,the similarity of samples from Guangdong Province was over a wide range of 0. 97- 0. 78,and the similarity of samples from Hainan Province was in 0. 88 range. CONCLUTION The established fingerprint method can effectively distinguish cuttlebones from other marine shells.
