摘  要:目的:研究牛樟芝提取物(ANCA-E-D)抑制人肝癌细胞SMMC-7721增殖的作用,揭示其可能机制。方法:取对数生长期人肝癌细胞SMMC-7721,以6×104/m L接种于96孔板,每孔100μL,随机分为调零组、空白对照组、溶剂对照组以及20,40,60 mg·L-1ANCA-E-D药物组,每组设5个复孔,分别作用24,48,72 h,采用MTT比色法检测ANCA-E-D对SMMC-7721细胞增殖的影响;通过瑞氏染色观察以上浓度的ANCA-E-D对肝癌细胞SMMC-7721形态学的影响;通过流式细胞仪检测以上浓度的ANCA-E-D对SMMC-7721细胞周期和早期凋亡的影响。结果:MTT实验结果表明,20,40,60 mg·L-1的ANCA-E-D对人肝癌细胞SMMC-7721的增殖具有显著的抑制作用(P<0.05),呈时间和剂量依赖性,其半数致死率(IC50)分别为31.21,24.12,21.48 mg·L-1;形态学观测结果提示,ANCA-E-D药物组的细胞核碎裂、凋亡小体形成;流式细胞仪检测发现,不同浓度的ANCA-E-D作用后,SMMC-7721细胞凋亡以晚期凋亡为主,并呈剂量依赖性。结论:ANCA-E-D可抑制人肝癌细胞SMMC-7721的增殖,可能与其诱导SMMC-7721细胞凋亡有关。Objective: This study was aimed to investigate the inhibitory effect of extract of Antrodia camphorata(ANCA-E-D) in human hepatocellular carcinoma cells SMMC-7721, and to explore its possible mechanism. Method: In this study,we used the exponential phase cells. The MTT method was used to detected the inhibitory effect of ANCA-E-D in various concernation on proliferation of SMMC-7721 cells which were cultured in vitro after 24,48,72 hours. Cell morphological changes were observed after Wright's stain. The flow cytometry was adopted to detected the change of cell cycle and the detection of apoptosis in vitro. Result: The concentration of 20,40,60 mg·L- 1ANCA-E-D could markedly inhibit the proliferation of SMMC-7721 cells after 24,48,72 h. The 50% inhibiting concentration IC50 of SMMC-7721 cells for 24,48,72 hours were 31. 21,24. 12,21. 48mg·L- 1,respectively. The SMMC-7721 cells displayed distinctive apoptotic character by Wright's staining with ANCA-E-D. Flow cytometry indicated that the different concentrations of ANCA-E-D could induce SMMC-7721 cells apoptosis,that the late apoptosis is dominant. Conclusion: ANCA-E-D can inhibit the proliferation of human hepatocellular carcinoma cells SMMC-7721,and the possible mechanism may relate to the induction of apoptosis.
