化瘀涤痰汤对急性硬膜下血肿模型大鼠脑组织中HIF-1α和Claudin-5蛋白表达的影响

摘　　要：目的:探讨化瘀涤痰汤对急性硬膜下血肿大鼠脑组织中缺氧诱导因子1α(HIF-1α)和紧密连接蛋白5(Claudin-5)蛋白表达的影响。方法:复制急性硬膜下血肿大鼠模型,造模成功后将大鼠分为:假手术组、模型组、化瘀涤痰汤高剂量组(HG组)、化瘀涤痰汤中剂量组(HZ组)、化瘀涤痰汤低剂量组(HD组)、天丹通络胶囊阳性组(TD组),每组10只大鼠,每日给药1次,持续给药14天后处死取脑组织,免疫印迹法(Western Blot)检测大鼠血肿侧脑组织中HIF-1α和Claudin-5的蛋白表达量。结果:与假手术组比较,模型组大鼠注血侧脑组织中HIF-1α蛋白表达量上调(P<0.01),Claudin-5的蛋白表达量显著降低(P<0.05),与模型组比较,HG、HZ、HD组和TD组HIF-1α的表达均显著下调(P<0.05,P<0.01),HG组和TD组大鼠脑组织中Claudin-5表达均升高(P<0.01)。结论:化瘀涤痰汤降低急性硬膜下血肿大鼠脑组织中HIF-1α蛋白表达并增加Claudin-5蛋白的表达。Objective： To investigate the expression of HIF-1α and Tight junction protein-5（Claudin-5） in the brain tissue of rat with subdural hematoma. Method： The rat model of acute subdural hematoma in duplicate, after the success of Mod- eling Rats shorthand is divided into： Control group n=10, Model group n=10, High dose group（Group HG, n = 10）, Middle dose group （Group HZ, n = 10）, Low dose group （Group HD, n = 10）, Positive group of Tiandan capsule（group TD, n= 10）, onlyl time daily administration. At the 14th day, the rats were sacrificed to detect the expression of protein HIF-1 α and Claudin-5 by WB. Result： Compared with the Control group, in the blood injection side of the model rats brain tissues, ex- pression of HIF-1 α were raised（P〈0.01）, significantly the amount of Claudin-5 protein expression was reduced（P〈0.05）; compared with the Model group, HIF-1α expression of HG, HZ, HD and TD groups were significantly lowered（P〈0.01, P〈0.05）, Claudin-5 expression of HG and TD groups were increased（P〈0.01）. Conclusion： Huayu Ditan decoction to play a protective role by reducing HIF-1α protein expression and increased Claudin-5 protein expression in brain tissues of rats with acute subdural hematoma.
