摘  要:目的:观察加味四君子汤合骨髓间充质干细胞(BMSCs)对阿霉素肾病大鼠骨代谢的影响。方法:制作大鼠阿霉素肾病模型,将60只SD大鼠随机分为6组,即A组:正常组;B组:模型组;C组:激素组;D组:干细胞组;E组:干细胞+激素组;F组:中药+干细胞组。各组于造模后第7天、第21天和第35天收集24h尿标本,以双缩脲比色法测定大鼠尿蛋白,以ELISA法测定血清骨保护素(OPG)、核因子κB受体活化因子配体(RANKL)水平,用实时定量PCR检测骨组织中OPG、RANKL及黏着斑激酶(FAK)的表达,Western blot检测骨组织FAK的表达。结果:与A组比较,第7天,各组尿蛋白升高;与B组比,第21天和35天,C、D、E、F组尿蛋白降低。与B组比较,C组血清OPG、OPG m RNA和FAK m RNA及其蛋白均降低,血清RANKL和RANKL m RNA均升高。与C组比较,D组、F组血清OPG及m RNA和FAK m RNA及其蛋白均升高,血清RANKL及m RNA均降低。与D组比较,E组血清OPG、OPG m RNA和FAK m RNA及其蛋白均降低,血清RANKL和RANKL m RNA均升高。结论:FAK-OPG/RANKL通路可能在泼尼松诱导的骨质疏松的形成中发挥着重要的作用,加味四君子汤合BMSCs能一定程度上逆转这一趋势而延缓诱导的骨质疏松的形成,且有协同作用。Objective: To observed the effects of Modified Sijunzitang combined bone marrow mesenchymal stem cells(BMSCs) on the bone metabolism of adriamycin-induced nephritic rats. Methods: Generate adriamycin-induced ne- phriticrats (n=60), and randomly divide them into six groups as group A:the normal group, group B: the model group, group C: the hormone group, group D: nephritic rats with stem cells transplant, group E: nephritic rats with stem cells transplant+ hormone treatment, group F: nephritic rats with stem cells transplant+TCM treatment.The urine samples were collected on the on the day 7, 21 and 35 respectively after adriamycin injection,and urine protein levels were measured by automatic biochemistry analyzer.The serum protein levels of osteoprotegerin(OPG), nuclear factor- K B receptor activat- ing factor ligand(RANKL) were detected by ELISA. Using real-time quantitative PCR to measure the expression of OPG, RANKL and FAK in tibia tissue.Using Western blot to measure the protein expression of FAK in tibia tissue. Results: com- pared to the group A, on the dayT, urinary protein level of group B to E all showed significant increase; compared to group B, day 21 and 35 day, the urinary protein of the C,D ,E and F decreased, in the group C, the serum of OPG, OPG mRNA and FAK mRNA with its protein decreased, the serum of RANKL, RANKL mRNA increased Compared to the B group; in both group D and F, the serum of OPG, OPG mRNA and FAK mRNA with its protein increased, the serum of RANKL, RANKL mRNA level were reduced as compared to the C group; Compared with the D group, E group showed decreased level of the OPG serum, mRNA with FAK mRNA, protein, and increased level of the RANKL serum and mRNA. Conclusion:FAK- OPG/RANKL pathway may play an important role in the formation of nrednisone-inducad ogtonnara^i~ wht^ron~ MnAir;,~,tSijunzitang combined BMSCs could slow the prednisone-induced bone oste〈〉porosis formation through reverse this trend to some extent, which can provide syne
