灵宝大枣无菌外植体的建立及茎段不定芽的诱导

摘  要:为建立灵宝大枣的快速繁殖体系,以灵宝大枣枣头一次枝上萌发的嫩枝为外植体,对外植体的消毒、茎尖的生长和茎段不定芽的诱导进行了研究。结果表明:灵宝大枣外植体采用乙醇和升汞结合的常规消毒方法较好,污染率较低;适宜茎尖生长的培养基为MS+6-BA0.5mg·L-1+NAA0.1mg·L-1+AC0.3%+蔗糖3%+琼脂粉7.0g·L-1,培养基MS+TDZ0.5 mg·L-1+NAA0.2 mg·L-1和MS+6-BA0.5 mg·L-1+NAA0.1mg·L-1+AC0.3%适宜枣茎段不定芽诱导,活性炭可以明显减轻褐变,TDZ诱导茎段不定芽的能力明显高于6-BA。In order to establish rapid proliferation for Lingbao jujube, taking sprout shoots from jujube first branch as the explants,the disinfection of explant,the growth of stem tip and adventitious bud induction from stem were studied. The results showed that the combination of 75% alcohol and 0.1% HgCI2 had better effect of disinfecting explants, and the pollution rate was low. The suitable medium for stem tip was MS+6-BA 0.5 mg.L-1I+NAA0. 1 mg-L-1 +0. 3%AC+3% Sugar+Agar 7. 0 g-L-1,the mediums of MS+TDZ 0.5 mg.L-1 +NAA0.2 mg-L-1 and MS +6-BAO. 5 mg.L-1 + NAA0.1 mg.L-1 +0.3 %AC were suitable for inducing adventitious buds, activated carbon added to medium could reduce browning significantly, the ability of inducing adventitious bud of TDZ was higher than that of 6-BA obviously.
