摘  要:目的:研究当归芍药散不同提取部位对体外培养的大鼠海马神经干细胞的增殖作用。方法:以无血清传代培养法,从胎鼠中获得神经干细胞;传至3代后用免疫荧光法对神经干细胞进行鉴定;将当归芍药散不同部位的含药培养基加入神经细胞中进行培养,于第10天进行免疫细胞化学法染色,观察神经干细胞增殖标志物Brdu的表达。结果:当归芍药散的醇提部位(浓度为50μg/ml,100μg/ml)、二氯甲烷部位(浓度为100μg/ml)对体外培养的神经干细胞有明显的增殖作用(P<0.05),正丁醇部位和剩余水部位对体外培养的神经干细胞无明显增殖作用。结论:当归芍药散的醇提部位和二氯甲烷部位可显著促进体外培养大鼠海马神经干细胞的增殖。Objective: To investigate the effects of four fractions of Dangguishaoyaosan on the proliferative activities of neural stem cells( NSCs) in vitro. Method: NSCs were isolated from 16 d SD rats and cultured in serum-free medium. After the third passage,NSCs were detected by the immunofluorescence technique with nestin antibody. NSCs were co-incubated with the different fractions of Dangguishaoyaosan and immunofluorescence staining was used to observe the proliferation potential. Results: Fractions of Et OH( at the concentrations of 50 μg / ml and100 μg / ml) and CH2Cl2( at the concentration of 100 μg/ml) of Dangguishaoyaosan could significantly promoted the proliferation of neural stem cells,and fraction of n-butnol and H2 O had no obvious effect on the proliferation. Conclusion: Fractions of Et OH and CH2Cl2 of Dangguishaoyaosan can significantly enhance the proliferative activities of neural stem cells in vitro.
