雷公藤甲素致大鼠颗粒细胞凋亡及雌激素保护作用研究

摘　　要：目的:探讨雷公藤甲素(triptolide,雷公藤甲素)致雌性大鼠生殖系统损害及防治方法。方法:采用体外卵巢颗粒细胞(granulosa cell,GCs)培养,分别给予不同浓度雷公藤甲素干预及不同浓度β-雌二醇与雷公藤甲素共同孵育,观察卵巢GCs的生长情况,采用MTT检测细胞增殖程度,TUNEL检测细胞凋亡程度。结果:细胞培养48小时后观察各组细胞形态,雷公藤甲素浓度大于100ug/L时,贴壁细胞数量明显减少;MTT检测显示,与对照组相比,当浓度>100ug/L的雷公藤甲素组均存在显著差异。TUNEL检测细胞凋亡提示β-雌二醇(10-7mol/L)与雷公藤甲素(2.78×10-7mol/L)共同孵育组,细胞凋亡程度明显减少,数量增多。结论:随着雷公藤甲素浓度增加,颗粒细胞生长及增殖受到显著抑制;β-雌二醇可以预防和修复雷公藤甲素对大鼠颗粒细胞造成的伤害,增强细胞活性,抑制细胞凋亡。Objective ：To explore triperygium （TP） -induced injury of female rats genital system, and explore the protective effects of estrogen on genital system. Methods： The granulosa cell（GC） from the ovary of rats was cultured in vitro and exposed to various concentrations of trip- tolide and estrogen. MTT was used to examine the effects of triptolide on proliferation of cells ,TUNEL was used to detect apoptotie cells. Re- sult： To observe the GC morphology of different concentrations of TP after 48h. When the concentration of TP was higher than 100ug/L, the number of the adherent cells was significandy reduced. Result of MTT shows that TP higher than 100ug/L can significantly inhibit the cell proliferation compared with the control group. The inhibitive effect was related to the drug concentration（ P 〈 0.01 ）. Result of TUNEL shows the degree of apoptosis on the group of estrogen（10-7mol/L） combined with TP （100ug/L）was significantly reduced. Conclusion： Different concentrations of TP could significantly inhibit the proliferation and the inhibition enhanced with the increasing of concentration of TP, with a dose-dependent relationship. Estrogen could effectively repair the damage of GCs caused by TP, enhance the activity of GCs, inhibit the degree of apoptosis.
