摘  要:目的:观察黄精多糖对2型糖尿病大鼠肝脏的保护作用及SREBP-1c蛋白表达情况。方法:取80只雄性大鼠高脂高糖饲料喂养2个月后,腹腔注射链脲佐菌素(STZ)25 mg/kg制备糖尿病大鼠模型,模型成功后随机分组为:正常对照组、黄精多糖(480、240、120mg/kg)组、二甲双胍(150 mg/kg)组,各治疗组分别灌胃给予相应药物,正常组和模型组给予生理盐水,共8周实验结束后测定空腹胰岛素(FINS)、血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)和游离脂肪酸(FFAs);HE染色后光镜观察肝脏病理变化;Western Blot法检测SREBP-1c和SCD-1蛋白表达水平。结果:与模型组比较,黄精多糖480 mg/kg、240mg/kg剂量组FBG、TG、TC、LDL-C、FFAs水平均明显降低,HDL-C升高;HE染色显示各大鼠肝细胞脂肪变性明显减少,肝细胞排列基本规则;Western Blot结果显示SREBP-1c和SCD-1蛋白表达明显降低。结论:黄精多糖可降低糖尿病大鼠血糖、血脂,减轻肝细胞脂肪变性,其作用机制可能与降低SREBP-1c和SCD-1蛋白表达有关。Objective: To investigate the protective effects of Polygonatic Rhizome on liver in type 2 diabetic rats and the expression of SREBP-1c and SCD-1. Methods: Eighty male SD rats were fed high fat and sugar two months,after that they were injected dose of streptozotocin( STZ) to form diabetic model rats. Modeled rats were divided into normal,control group,and Polygonatic Rhizome groups( 480、240、120mg/kg),metformin group( 150 mg / kg). Each treatment group respectively by gastric was given corresponding drugs,the normal group and model control group was given physiological saline eight weeks,On the 8weekend,Blood lipid levels and fast insulin( FINS),serum triglyceride( TG),total cholesterol( TC),low density lipoprotein( LDL-C),high density lipoprotein( HDL-C) and free fatty acids( FFAs) were measured. The pathology changes of liver were observed in light-microscopic by HE staining. The expression level of SREBP-1c and SCD-1 was detected by Western Blot method. Results: Compared with the model group,the levels of FBG,TC,TG,LDL-C,FINS in 480 mg / kg、240mg /kg dose of Polygonatic Rhizome groups obviously decreased,but HDL-C significantly increased. HE staining showed that liver fatty degeneration was significantly reduced in Polygonatic Rhizome groups,liver cell arranged regularly. Western Blot showed that expression level of SREBP-1c was significantly reduced in Polygonatic Rhizome groups. Conclusion: These results suggest that Polygonatic Rhizome can significantly lower blood sugar and blood lipid,and alleviate liver fatty degeneration in diabetic model rats,which probably associated with the lowered expression of SREBP-1c and SCD-1.
