摘  要:目的:考察新疆软紫草石油醚提取物对小鼠的急性毒性和蓄积毒性影响,探讨其主要靶器官的毒性作用。方法:急性毒性实验:采用改良寇氏法进行半数致死量(LD50)的测定;蓄积毒性实验:采用剂量递增法给药,对照组给以4%吐温-80水溶液,观察实验期间一般生理情况,以及组织病理学变化,进行血清生化指标检测。结果:新疆软紫草石油醚提取物单次灌胃小鼠的半数致死量(LD50)=3.48g/kg,LD5095%的可信区间为3.46~3.50g/kg。蓄积毒性实验结果表明新疆软紫草石油醚提取物在小鼠体内的蓄积系数K=3.76,为中等蓄积毒性,小鼠17天累积灌胃新疆软紫草石油醚提取物13.08g/kg可引起小鼠摄食量减少,体重下降,小鼠肝脏、脾脏和胃肠道均有不同程度的损伤和病变。结论:新疆软紫草石油醚提取物对小鼠的毒性作用主要表现在肝毒性和胃肠道毒性,表明肝脏和胃肠道可能是其毒性作用的主要靶器官。Objective: To investigate the acute toxicity and cumulative toxicity of Arnebia euchroma petroleum ether extract effects in mice and discusses its main toxic target organs. Methods Acute toxicity: Using the impro ved karber method to determine the median lethal dose,The experimental group is divided into 6 groups and give different concentrations of Arnebia euchroma petroleum ether extract. The control group give 4% twain-80 aque ous solution. Cumulative tox icity: By using dose escalation for medicine,the control group give 4% twain-80 aq ueous solutio n. Usually the physiological information observed during the experiment and Histopathological cha nges. Detection of serum biochemical indexes in mice. Results: The Arnebia euchroma petroleum ether extract sin gle lavage LD50 in mice = 3. 48 g / kg. Accumulation toxicity experiment results show that the Arnebia euchroma petroleum ether extract medium accumulati on toxicity in mice and the accumulation coefficient K = 3. 76,which can cause food intake and weight loss in mice. Liver,spleen and gastro intestinal tract in mi ce have different deg rees of injury and disease. Conclusions: The toxic effects of Arnebia euchroma petroleum ether extract in mice in liver toxicity and gastrointestinal tract toxicity. This suggests that the liver and gastrointestinal tract is probably the main toxic target organs.
