摘  要:目的:对比葛根有效成分异黄酮和黄连有效成分总生物碱单用及配比后降糖作用及其血清化学成分差异。方法:灌胃给以葛根异黄酮800mg/kg、黄连总生物碱60mg/kg和葛根异黄酮黄连总生物碱8:3配比860mg/kg,比较灌胃葛根异黄酮组、黄连总生物碱组和葛根异黄酮黄连总生物碱配比组降糖作用;采用HPLC梯度洗脱方法对比葛根异黄酮和黄连总生物碱配比与含药血清化学成分的差异。结果:葛根异黄酮与黄连总生物碱单用及合用均能明显降低四氧嘧啶致高血糖大鼠血糖,改善血清甘油三酯(TG)和胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C),葛根异黄酮黄连总生物碱配比组优于葛根异黄酮组和黄连总生物碱组;葛根异黄酮黄连总生物碱配比组血清中有12个成分来源于原配比组,包括有葛根素、大豆苷、黄连碱、药根碱、巴马汀和小檗碱,有22个成分来源于代谢产物,其中6个代谢产物由葛根异黄酮代谢,5个代谢产物由黄连生物碱代谢,11个代谢产物来源于葛根异黄酮和黄连总生物碱的相互作用,同时2个原配比组中成分未在血清中检出。结论:葛根异黄酮与黄连总生物碱合用降糖效果优于其有效成分单用,原因可能因为主要有效成分入血后相互作用引起含量变化、新生有效代谢中间体造成。Objective: Compare Pueraria isoflavones and berberine total alkaloid use alone and proportion effecet on hypoglycemic,and serum chemical composition. Methods: Ig given isoflavones( G) 1000 mg / kg,total alkaloid berberine( H) 200 mg / kg,proportion isoflavones and alkaloid( GH proportion)( isoflavones: total alkaloids / 200mg/kg∶ 160mg/kg),compare hypoglycemic effect of gavage group G,group H and group GH ratio; using HPLC gradient elution method to compare the ratio soflavones and total alkaloid in the chemical composition of the drug in serum. Results: Isoflavones and total alkaloid berberine alone and in combination can significantly reduce diabetes rats fasting blood glucose induced by alloxan,improve serum triglyceride( TG) and cholesterol( TC),high density lipopmtein cholestero( HDL-C),low density lipopmtein cholesterol( LDL-C),GH ratio was superior to Group G and H group; GH serum has 12 ingredients from the prototype,including puerarin,daidzin,coptisine,jatrorrhizine,palmatine and berberine,22 ingredients derived metabolites,including 6 metabolites from the metabolism of isoflavones,five metabolites by berberine alkaloid metabolism,11 metabolites comes from the interaction of isoflavones and total alkaloid berberine arrowroot,while two components were not detected in the serum. Conclusion: Isoflavones combined with total alkaloid berberine hypoglycemic effect better than the active ingredient alone,probably because the main active ingredient into the blood caused by the interaction of content changes,effective newborn metabolic intermediate result.
