摘 要:目的:本研究旨在观察脂肪肝泰胶囊对氧四环素加高脂饲料致大鼠非酒精性脂肪肝的影响。方法:雄性大鼠88只,随机分为空白对照组和模型组,空白对照组喂以普通饲料,模型组以高脂饲料喂养,并每隔5天腹腔注射氧四环素1次,10mg/100g剂量。造模成功后将模型组分为5组:模型组、东宝肝泰组、脂肪肝泰胶囊0.74、1.48、2.96g(原生药)/kg三个剂量组,每组12只。连续给药4周后,处死动物,制备肝匀浆,检测、计算各组大鼠肝脏系数、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-C)、丙二醛(MDA)、超氧化物歧化酶(SOD)和肝组织病理切片。结果:与模型组相比,脂肪肝泰胶囊1.48、2.96g(原生药)/kg两个剂量组能明显改善非酒精性脂肪肝大鼠的肝细胞的脂肪病变程度,使脂滴减少,明显降低丙二醛(MDA)水平,使超氧化物歧化酶水平(SOD)升高;脂肪肝泰胶囊0.74、1.48、2.96g(原生药)/kg三个剂量组能明显降低肝脏系数,血清中胆固醇(TC)的含量分别降低了0.77、0.70、0.86mmol/L,而低密度脂蛋白(LDL-C)水平分别降低了0.15、0.11、0.15 mmol/L及肝组织中的甘油三酯(TG)含量分别降低了0.43、0.81、0.96mmol/L。结论:脂肪肝泰胶囊对大鼠非酒精性脂肪肝有明显对抗作用,其抗NAFLD的机制之一可能与其提高机体抗氧化能力有关。Objective: Based on the research and development of Curcuma which is a genuine medicinal material in Sichuan,Zhifanggantai capsule,a new drug of traditional Chinese medicine,is developed by Zhaojunning researcher of Academy of Chinese Medical Sciences in Sichuan province. The purpose of this research is to observe the effect of the drug on nonalcoholic fatty liver induced by oxytetracycline and high fat forage. Methods: 88 healthy male rats were randomly divided into control group and model group. Rats in the control group were fed with normal food-stuffs while those in the model group were fed with high fat food-stuffs and injected with oxytetracycline every five days. The dose was 10 mg / 100 g( per 1 ml ampoules containing 500 mg drugs). The model group were divided into five groups,every group containing 12 rats,including model group,dongbaogantai group and three zhifanggantai capsule groups( the primary drugs of each group are 0. 74,1. 48 and2. 96 g / kg),each group con-tains 12 rats after the model was developed successfully. They were administrated for four weeks successively,and then sacrificed to collect the blood and liver tissues wh-ich were used to determine the values of triglyceride( TG),total cho-lesterol( TC),low density lipoprotein( LDLC) level,malondialdehyde( MDA) and superoxide dis-mutase( SOD) activities respectively. Results: Zhifanggantai capsule( the primary drugs of each group are 1. 48 and 2.96 g / kg) could not only alleviate the depos-ition of fatty in the liver tissue and ameliorate the patholoical change of the hepatocytes significantly,but also reduce the activities of hepatical MDA significantly and increase activities of hepatical SOD; Zhifanggantai capsule( the primary drugs of each group are 0. 74,1. 48 and 2. 96 g / kg) could reduce the TC,LDL-C level of blood and TG of liver tissues sig-nificantly compared to model group.
