香芍胶囊对老龄雌鼠性激素及5-HT受体的影响

摘　　要：目的:研究香芍胶囊对老龄雌鼠下丘脑-垂体-卵巢轴相关激素、脏器系数及脑内5-HT受体表达的影响,探讨香芍胶囊对老龄雌鼠的作用。方法:以自然衰老的老龄雌鼠为动物模型,香芍胶囊2.75、5.50、11.00g(原生药)/kg连续给药8周,采用放射免疫法测定血清中雌二醇(E2)、促黄体生成素(LH)、促卵泡激素(FSH)、泌乳素(PRL)、孕酮(P)的含量,用Elisa试剂盒测定血清中促性腺激素释放激素(GnRH)的含量,并用免疫组化方法观察大鼠下丘脑及海马中5-HT受体表达的变化。结果:香芍胶囊11.00g/kg组能提高E2、PRL水平;香芍胶囊11.00、5.50g/kg组可升高血清中P、GnRH水平;香芍胶囊2.75g/kg组能提高子宫系数;香芍胶囊11.00g/kg组能提高胸腺系数;香芍胶囊11.00g(原生药)/kg组能提高大鼠下丘脑中5-HT受体的阳性表达水平(P<0.05);香芍胶囊11.00、5.50g(原生药)/kg组可提高大鼠海马中5-HT受体的阳性表达水平。结论:香芍胶囊对老龄雌鼠下丘脑-垂体-卵巢轴相关激素有调节作用,能够提高血清中E2、P、PRL、GnRH水平,香芍胶囊能提高子宫系数和胸腺系数,能改善老龄雌鼠的易惊慌易激惹等行为表现,香芍胶囊对更年期综合征治疗作用可能与其对下丘脑-垂体-卵巢轴的调节和对下丘脑和海马中5-HT受体表达的提高有关。Objective： To research the effect on Xiangshao capsules for hypothalamic-pituitary-ovarian axis and organ coefficients and 5-HT receptor expression in the brain,to explore the mechanism of Xiangshao capsules about older female rats. Method： Natural aging older female rats was used for animal model. 2. 75,5. 50,11. 00 g / kg of Xiangshao capsules was continuous administrated for 8 weeks. Determined estradiol（ E2）,luteinizing hormone（ LH）,follicle-stimulating hormone（ FSH）,prolactin（ PRL）,progesterone（ P） of serum by radioimmunoassay.Gonadotropin-releasing hormone（ GnRH） was determined with Elisa Kit. 5-HT receptor expression in the thalamus and hippocampus was observed by immunohistochemical. Results： The lever of E2,PRL was increased by 11. 00g/kg of Xiangshao capsules（ P〈0. 05、P〈0. 01）.The lever of P,GnRH was improved by 11. 00,5. 50 g / kg of Xiangshao capsules（ P〈0. 01）. 2. 75 g / kg of Xiangshao capsules Increased uterine factor（ P〈0. 05）. The thymus factor was improved by 11. 00 g / kg of Xiangshao capsules（ P〈0. 05）. The level of 5-HT receptors expression in rat hypothalamus was increased by 11. 00 g / kg of Xiangshao capsules（ P〈0. 05）. The level of 5-HT receptors expression in rat hippocampus was increased by 11. 00 g / kg of Xiangshao capsules（ P〈0. 05）. Conclusion： The related hormones of hypothalamic-pituitary-ovarian axis was regulated in older female rats by Xiangshao capsules. It increased the lever of E2,P,PRL,GnRH and improved uterine factor and thymus factor. It improved the behavior of easy panic and irritability of older female rats. Xiangshao capsules had treatment action about menopausal syndrome,its main mechanism of action may be related to the regulation of hypothalamus-pituitary-ovarian axis and improvement of 5-HT receptors expression in thalamus and hippocampus.
