摘 要:制备了新型多壁碳纳米管-纳米金-壳聚糖复合材料修饰的丝网印刷电极(SPE),研究了氨茶碱在该电极上的电化学行为,并优化了缓冲液pH、富集时间、扫描频率等测定条件。在最适条件下,以方波伏安法(SWV)测得氨茶碱在1.0V左右有灵敏的氧化峰,氧化峰电流大小和氨茶碱浓度在10~200μmol/L范围呈良好的线性关系,检出限为1μmol/L。标准曲线:Ip(μA)=0.0419c(μmol/L)+0.6303,R=0.9954。本实验对不同给药时间小鼠的氨茶碱血药浓度进行了测定,并与高效液相色谱法(HPLC)测定结果进行比较。本方法修饰得到的丝网印刷电极系统应可用于实际血样中氨茶碱血药浓度的测定。A novel CS/AuNPs-MWNTs modified screen printed electrode (SPE) was fabricated for directly simuhaneous determination of aminophylline (AMI) in whole blood sample. Electrochemical performance of the modified electrode was greatly enhanced with optimized experimental conditions. The currents of AMI measured by SWV presented a good linear property with the increace of the concentration. Under optimized conditions, the linear range of AMI was determined to be 10 ~ 200 μmol/L, with the detection limit of 1 μmol/L and peak potential of 1.0V, respectively. The equation of the linear regression was Ip (μA) = 0. 0419 c (txmol/L) + 0. 6303, which met the actual needs of detection of the plasma AMI concentration with a linear correlation coefficient of 0. 9954. Compared to the results detected by high performance liquid chromatography ( HPLC ), the proposed method appeared easier and more accurate. Therefore, the CS/AuNPs-MWNTs modified screen printed electrode (SPE) may offex an easy, quick and accurate electrochemical method to detect the concentration of AMI in the blood.
