生物与化学方法相结合提高海洋生境放线菌YB-24菌株星形孢菌素产量

摘　　要：分离自北极沉积物的海洋链霉菌Streptomyces sp.YB-24菌株产生强抗肿瘤活性的吲哚咔唑生物碱。为了提高其产量,先经过紫外线与氯化锂复合诱变,再通过抗真菌抗肿瘤稻瘟酶模型初筛与高效液相色谱HPLC检测跟踪相结合,获得的高产菌株,其代谢星形孢菌素的能力为3.833mg/L,比原始菌株提高342.28%。通过改变碳氮源及培养配方,在特定时期加入前体物色氨酸的方法,证明淀粉与磷酸氢二铵是菌株最合适的碳氮源,在起始期添加色氨酸时,产量为对照的216.40%,提高较明显。The marine Streptomyces YB-24 isolated from north pole produces indolocarbazole alkaloids with strong anti-tumor activity.In order to improve its production and its homologues amounts,YB-24 was treated with ultraviolet-LiC1, and then detected the production of staurosporine by HPLC method, a mutant strain L25-1 was obtained after a compound mutation, whose production was raised to 342.28%, compared with the initial strain. By changing the carbon, nitrogen sources, culture media and adding tryptophan in particurlar period, the results showed that starch and diammonium phosphate is the most suitable carbon and nitrogen sources, furthermore, adding tryptophan in initial period the production of staurosporine was raised to 216.40%, compared with the initial strain, the production of staurosporine in the medium ISP5 was significantly higher comt^ared with other mediums.
