摘  要:目的采用HPLC-电喷雾检测器(CAD)新技术测定硫酸阿米卡星中卡那霉素和硫酸盐的含量。方法采用华谱Click XIon(4.6mm×250mm,5μm)柱,流动相为含0.7%甲酸的250mmol/L甲酸铵溶液-乙腈-水(26:35:39),流速为1.0m L/min。采用Conona ultra RS CAD检测器检测,喷雾器温度为25℃。结果卡那霉素、硫酸盐与阿米卡星、中间体、副产物、辅料完全分离。卡那霉素、硫酸盐在一定的浓度范围内呈较好的线性关系,卡那霉素测定的回收率为100.7%(RSD=2.00%,n=9);硫酸盐测定的回收率为100.2%(RSD=1.04%,n=9)。结论方法简便、灵敏、重复性好,可用于本品的质量控制。Objective A new HPLC- charged aerosol detector technology for the determination of kanamycin and sulfate in amikacin sulfate was developed. Methods The column was Acchrom Click XIon (4.6mm×250mm, 5μm). The mobile phase was 250mmol/L ammonium formate and 0.7% formic acid aqueous solution-acetonitrile- water(26:35:39); the flow rate was 1.0mL/min. Detector was an Conona ultra RSCAD detector. The sprayer temperature was 25℃ Results Good separation of kanamycin and sulfate from amikacin and main intermediates could be achieved. The standard curves of kanamycin and sulfate were rectilinear in the certain range. The average recovery of kanamycin is 100.7%(RSD=2.00%, n=9). The average recovery of sulfate is 100.2%(RSD=1.04%, n=9). Conclusion The method is simple, sensitive and reproducible, and can be used for quality control of amikacin sulfate.
