摘 要:目的:观察菟丝子含药血清干预肿瘤坏死因子α(TNF-α)诱导建立人蜕膜细胞异常凋亡模型后基因表达谱的变化,以期从基因表达谱的变化探讨菟丝子安胎的分子机理。方法:体外常规进行人蜕膜细胞的培养,选用适宜浓度的TNF-α诱导建立人蜕膜细胞异常凋亡模型,用菟丝子含药血清干预,采用一步法抽提RNA,用人全基因组17Kc DNA基因芯片来分析菟丝子作用后的基因表达谱变化情况,分析基因的变化趋势。结果:菟丝子含药血清作用于TNF-α诱导的异常凋亡的蜕膜细胞后共检出基因10 230条,其中共检出表达差异基因128条,上调基因76条,下调基因52条;这些表达差异基因有细胞因子及受体、转录相关基因、凋亡相关基因、生长分化相关基因、信号转导相关基因、细胞周期相关基因、电子传递和氧化相关基因、转移酶相关基因、核酸酶相关基因等。结论:菟丝子可能通过以上基因的表达,调控蜕膜细胞的生长周期及异常凋亡,在免疫调节等方面发挥有一定的安胎作用。Objective:To study the changes in gene expression profiles after establishing the abnormal apoptosis model of human decidual cells induced by intervention of dodder medicated serum on TNF- alpha gene, and so as to discuss the the molecular mechanism on tocolysis of dodder from the changes of gene expression profile. Methods: The human decidual cells was cultured in vitro. The abnormal apoptosis model of human decidual cells,which was established induced by TNF-α at appropriate concentration,was intervened by dodder medicated serum. RNA was extracted by one footwork method. The changes of gene expression profile after the action of dodder was analyzed by entire genome team 17KcDNA gene chip to analyze the varying tendency of the gene. Result: 10,230 genes were detected after abnormal apoptosis of human decidual cells induced by intervention of dodder medicated serum on TNF-α, 128 of differentially expressed genes,and 76 of up-regulated genes, 52 of down-regulated genes. These dif- ferentially expressed genes had cytokines and their receptors, gene transcription,apoptosis-related genes, growth and differentiation-related genes, signal transduction genes, cell cycle-related genes, electron transport and oxidative-re- lated genes,metastasis-associated gene,nuclease-related genes and so on. Conclusion: Dodder can regulate growth cycle and abnormal apoptosis of decidual cells, and affect the secretion of cytokines, and play a certain role on tocol- ysis in immune regulation through the expression of the above genes.
