摘 要:目的:对DiI标记固定大脑皮质内神经元及其突起的技术进行改进,使操作更简单、便捷,效果更为高效、理想.方法:将DiI颗粒植入已固定小鼠大脑胼胝体,然后将标记后的脑组织放入4%多聚甲醛固定,于室温下避光孵育1~2周.结果:这种标记方法可以很直观地显示出大脑皮质神经元及其突起的精细结构,其中包括树突棘.结论:DiI示踪标记法可以用于标记固定脑内的神经元及其突起和树突棘,效果令人满意.Objective: To modify the DiI tracing method to label neurons and their processes in the fixed mouse brain. Methods: After two hemispheres were separated, DiI was implanted into the corpus callosum of fixed mouse brain, and then the brains were post-fixed in 4% paraformaldehyde at room temperature and kept in dark for 1-2 weeks. Results: The fine structure of pyramidal cells and their processes including dendritic spines could be visualized well with this method. Conclusion: Our results suggests that the DiI tracing method could be used to label the neuronal cell bodies, processes and dendritic spines in the fixed brain satisfactorily.
