摘 要:本研究利用分布于水稻12条染色体上的12个SSR标记构建了太湖稻区10份抗褐飞虱粳稻的DNA指纹图谱,并对粳稻品种之间的遗传相似性进行了分析,旨在为水稻抗性育种、种子纯度鉴定和新品种知识产权保护方面提供支持。研究结果显示:12对引物共检测到35个等位基因,每对SSR引物检测到等位基因为1~6个,平均为2.8个;筛选出3对核心引物(RM336, RM5414, RM190)并建立了10份抗褐飞虱粳稻的SSR指纹图谱;10份粳稻品种之间的相似系数变幅为0.20~0.91,平均为0.63,遗传相似系数在0.50以上的材料占全部的82.22%。UPGMA聚类结果显示:在遗传相似系数0.55处,10份粳稻材料可以分为2个类群。研究结果表明,太湖地区绝大多数抗褐飞虱粳稻品种遗传相似性高,遗传多样性不够丰富。DNA fingerprint map of japonica rice (Oryza Satiua L. Subsp. japonica) with resistance to brown plan-thopper in Taihu area was constructed and genetic similarity of them was analyzed by simple sequence repeat (SSR) to provide support for rice resistance breeding, seed purity identification, intellectual-property protection. A total of 12 SSR markers distributed in 12 chromosomes were use. As a result, a total of 35 loci were detected among 10 varieties using the 12 SSR primers. The average number of detected loci each primer pair was 2.8 with a range from 1 to 6. In this study, 3 pairs of core SSR makers (RM336, RM5414 and RM190) were screened among 12 pairs of SSR primers to establish DNA fingerprinting of the japonica rice with resistance to brown planthopper. The genetic similarity coefficient among 10 japonica rice varieties ranged from 0.20 to 0.91, with average 0.63, and the genetic similarity coefficient of 82.22%tested materials is 0.50 or more. Furthermore, the 10 japonica rice varieties were classified into 2 groups, at the level of genetic similarity coefficient 0.55 according to the results of UPGMA. The results showed that the genetic background comparability of majority japonica rice varieties with resistance to brown planthopper in Taihu was high but the diversity was narrow.
