摘 要:目的探讨黄芪对创伤性脑损伤后紧密连接相关蛋白(claudin-5)表达和血脑屏障通透性的影响。方法 70只SD大鼠按随机数字表法分为对照组和黄芪组各35只,两组各30只采用自由落体致伤原理造模成功。黄芪组在制作大鼠脑损伤模型前7 d至处死时,每隔12 h腹腔注射黄芪,对照组用等量生理盐水腹腔注射。两组分别于大鼠脑损伤模型制成后6 h、1 d、3 d、5 d、7 d各取2只大鼠,用Western Blot法测定脑组织claudin-5的表达,采用伊文蓝(EB)法测定血脑屏障通透性。结果脑损伤后各个时间点黄芪组脑组织claudin-5蛋白表达量均明显高于对照组(P<0.05),脑组织EB含量均明显低于对照组(P<0.05)。结论调节claudin-5的表达,进而调节血脑屏障的通透性可能是黄芪抑制脑水肿形成的机制。Objective To investigate the effect of astragalus on tight junction-associated protein (clandin-5) expression and blood-brain barrier permeability in rats with traumatic brain injury. Methods Seventy SD rats were divided into control group ( n = 35 ) and astragalus group (n = 35) according to the random number table. The models of brain injury were established according to the principle of free falling injury,with 30 in each group. The rats in the astragalus group received the intraperitoneal injection of astragalus every 12 hours,7 days before the rat model of brain injury was established, and astragalus was replaced by equivalent volume of normal saline in the control group. The expression of claudin-5 in brain tissues was detected by Evans blue (EB) assay and the permeability of blood-brain barrier was determined by Western Blot in two groups individually 6 hours,1,3 ,5 and 7 days(2 rats per group at each time point) after the rat model of brain injury had been established. Results The expression of clandin-5 protein in brain tissues increased significantly while the content of EB in brain tissues decreased significantly in the astragalus group at all time points after traumatic brain injury compared with control group ( P 〈 0.05 ). Conclusion Astragalus regulates the expression of claudin-5 and the permeability of blood-brain barrier, which might be the mechanism of astragalus' inhibition on the formation of brain edema.
