摘  要:目的探讨牛蒡子苷对于人舌鳞状细胞癌Tca8113细胞增殖的调节作用及相关的分子机制。方法将Tca8113细胞分为对照组(Control组)、牛蒡苷干预组(ATG干预组)及牛蒡苷+p38 MAPK抑制剂SB203580组(ATG+SB组)3组。使用MTT法对不同时间点(0h,24h和72h)各组Tca8113细胞活性进行检测;使用western blot法对干预24小时后Tca8113细胞的p38 MAPK的活化水平及caspase-3的活化水平进行分析。结果与对照组相比,给予牛蒡苷干预的Tca8113细胞活性呈时间依赖性下降,差异均有统计学意义(P均<0.05),且各时间点ATG组较ATG+SB组细胞活性下降程度更加明显,差异均有统计学意义(P均<0.05)。干预24小时后,给予牛蒡苷干预的Tca8113细胞p38MAPK及caspase-3的活化水平均有显著的上升,差异有统计学意义(P均<0.05);但ATG组较ATG+SB组细胞p38 MAPK及caspase-3的活化水平上升更加明显,差异有统计学意义(P均<0.05)。结论牛蒡苷可以通过激活p38 MAPK促进人舌鳞状细胞癌Tca8113细胞caspase-3的活化,从而抑制Tca8113细胞的增殖。本实验为牛蒡子苷应用于舌癌和其他肿瘤的临床治疗奠定了一定的基础。Objective To investigate the inhibitory effect of arctigenin on the proliferation of tongue cancer Tea8113 cells and its potential mechanism. Methods Tca8113 cells were divided into 3 groups including control group, arctigenin (ATG) group and arctigenin+SB203580 group (ATG+SB) group. MTT was used to analyze the cell viabili- ties of Tca8113 cells in different time points at Oh, 24h and 72h. 24 hours after interventions, the activation of p38 MAPK and caspase-3 were measured by Western blot. Results The cell viabilities of Tca8113 cells were time-dependently reduced by ATG treatment. However, SB203580 blunted this effect of ATG. 24 hours after intervention, the activation of p38 MAPK and caspase-3 were noticeably increased by ATG. Meanwhile, we also found that these effects of ATG were largely abrogated by SB203580. Conclusion The proliferation were inhibited by arctigenin (ATG) through activa- tion of p38 MAPK/caspase-3 pathway in tongue cancer Tca8113 cells.
