摘 要:目的:探讨黄连解毒汤对2型糖尿病(T2DM)大鼠认知功能及脑内Alzheimer病样tau蛋白磷酸化途径的影响。方法:健康雄性Wistar大鼠72只,随机分为6组,分别为空白组、模型组、罗格列酮组、黄连解毒汤低、中、高剂量组(1.5、3、6g/kg)。通过高糖高脂饮食及小剂量链脲佐菌素(STZ)制备T2DM模型。除空白组、模型组给予等容积的蒸馏水,其他治疗组以相应药物治疗。8周后,采用Morris水迷宫观察大鼠学习记忆能力,Western blotting法检测大鼠大脑皮层tau蛋白在不同位点的磷酸化水平。结果:黄连解毒汤可缩短T2DM模型大鼠平均逃避潜伏期(P<0.01),增加逃逸平台次数(P<0.05)、延长有效区停留时间及有效区游泳路程(P<0.05,P<0.01),有效抑制模型大鼠脑组织tau蛋白在Ser199、Ser202、Thr231、Ser396等多位点的磷酸化程度(P<0.05,P<0.01)。结论:黄连解毒汤能够有效改善T2DM模型大鼠学习和记忆能力,可在tau蛋白多位点上抑制其过度磷酸化水平。Objective: To investigate the effects of Huanglian Jiedu Decoction on learning and memory ability and tan protein phosphorylation pathway of Alzheimer-like in rats with type 2 diabetes. Methods: Seventy-two healthy male Wistar rats were randomly divided into six groups, such as control group, model group, Rosiglitazone group, high dosage of Huanglian Jiedu group (6g/kg, HHJG), middle dosage of Huanglian Jiedu group (3g/kg, MHJG) and low dosage of Huanglian Jiedu group (1.5g/kg, LHJG). Type 2 diabetes rat models were established by feeding high sugar and high fat diet and injecting with low dosage of Streptozocin. Rats in control group and model group received garage administration with distilled water, and rats in the other groups received gavage administration with corresponding medicines, and all the treatment lasted for 8 weeks. Learning and memory ability of rats was observed by using Morris water maze. Western blotting was used to test the phosphorylation level of tau protein on different sites in rat cerebral cortex. Results: Huanglian Jiedu Decoction could shorten the escape latency of rats with type 2 diabetes (P〈0.01), increase the number of escape platform (P〈0.05), prolong residence time and distance swan in effective area (P〈0.05, P〈0.01), and inhibit phosphorylation extent of tau protein on different sites as Ser199, Ser202, Thr231 and Ser396 in rat brain effectively (P〈0.05, P〈0.01). Conclusion: Huanglian Jiedu Decoction could improve the learning and memory ability of rats with type 2 diabetes and inhibit the phosphorylation extent of tau protein on different sites.
