摘 要:目的:观察产后抑郁症(PPD)模型大鼠Treg和Th17细胞比例的变化,从T细胞免疫入手,探讨参芪解郁方调节PPD Treg/Th17平衡的作用机制。方法:90只SD雌性大鼠随机分为正常组、假手术、模型组、中药组和西药组,每组6只,分设灌胃1、2、4周3个时点。采用流式细胞术检测3个时点各组大鼠外周血Treg细胞和Th17细胞比例,并计算Treg/Th17比值。结果:各时点模型组Treg细胞数量较正常组降低,1周和2周较同时点下降明显(P<0.05,P<0.01);1周、2周模型组Th17细胞数量较正常组明显下降(P<0.01),而4周模型组Th17细胞数量较正常组明显上升(P<0.01),2周西药组较模型组升高明显(P<0.01),4周中药组较模型组降低明显(P<0.01);1周、2周模型组Treg/Th17较正常组明显升高(P<0.01),而4周模型组Treg/Th17较正常组明显降低(P<0.01),4周中药组较模型组升高明显(P<0.01)。结论:Treg/Th17平衡参与PPD发生发展,参芪解郁方通过调控Treg/Th17的失衡状态发挥效用,优于西药。Objective: To observe the change of proportion of Treg and Th17 cells in postpartum depression (PPD) rats, and to explore the mechanism of Shenqi Jieyu Formula (SJF) on regulating the balance of PPD Treg/Th17 from T cell immunity. Methods: Ninety female SD rats were divided into five groups, namely normal group, sham group, model group, SJF group and medicine group, 6 rats in each one, and the rats were gavage administrated for 1, 2, and 4 weeks. The proportion of Treg and Thl7 cells was observed in peripheral blood mononuclear cells of each group with flow cytometry in 3 time points, and the ratio of Treg/Thl7 was calculated. Results: Treg cells in the model group were lower than that in the normal group in each time point, significantly in 1-week and 2-week time point (P〈0.05, P〈0.01); Th17 cells in the model group were lower in 1-week and 2-week time point than that in the normal group (P〈0.01), while that were increased in 4-week time point (P〈0.01); Th17 cells were increased in the medicine group in 2-week time point when compared with that in the model group (P〈0.01), while in the SJF group in 4-week time point were decreased (P〈0.01). When compared with that in the normal group, the Treg/Th17 was significantly increased in 1-week and 2-week time point in the model group (P〈0.01), while apparently decreased in 4-week time point (P〈0.01), which in the SJF group were increased than that in the model group (P〈0.01). Conclusion: Imbalance of Treg/ Th17 may be a potent pathogenesis of PPD, and SJ Fang could be an effective therapeutic for regulating Treg cells and Thl7 cells in PPD.
