摘　　要：目的:探讨雌激素受体(ER)β基因启动子区Cp G岛甲基化状况在早发冠心病(PCHD)血瘀证发生发展中的作用及其临床意义。方法:收集早发冠心病血瘀证标本16例,冠心病非血瘀证(以痰浊证为例)8例,正常健康人8例,采用重亚硫酸盐纯化(BSP)和甲基化特异性PCR(MSP)技术对上述基因Cp G岛甲基化进行检测,比较并分析。结果:3组病例在年龄、性别、血脂、血压等方面不存在显著性差异,具有可比性。血瘀证组的基因甲基化程度为51.7%,大于非血瘀证组34.6%和健康人组12.8%,其差异具有显著性(P<0.05)。结论:ER-β基因甲基化可能与PCHD血瘀证的发生发展具有一定的相关性,检测其基因的甲基化状况可能对PCHD血瘀证的诊断具有一定的临床指导意义。Objective： In this paper, we focused on the clinical significance of estrogen receptor gene β methylation abnormalities in the occurrence and development process of blood stasis syndrome （bss） of premature coronary heart disease （PCHD）. Methods： We collected samples of bss of PCHD 16 cases, CHD non-blood stasis （non-bss, taking phlegm syndrome as an example） 8 cases, healthy people 8 cases. Applying Bisulfite Purified （BSP） and methylation-specific PCR （MSP） technology to detect, compare and analyze CpG island methylation of this gene. Results： Three groups of patients had no significant difference in age, sex, blood lipids, blood pressure and other aspects, they were comparable. BSS group ER-β gene methylation （51.7%） was higher than that of Non-BSS group （34.6%） and the healthy control group （12.8%） （P〈0.05）. Conclusion： ER-β gene methylation may have some correlation with occurrence and development in bss of PCHD, detecting the methylafion status of this gene may have some clinical significance in diagnosis of bss of PCHD.
