摘 要:目的:研究甘遂-甘草反药组合的量-毒关系,探讨甘遂与甘草配伍的毒性变化特点,为揭示甘遂"反"甘草提供科学证据。方法:ICR小鼠随机分为空白对照组、不同剂量甘遂及甘草单味药组、甘遂-甘草不同配伍比例给药组,共27组。在较大剂量范围(甘遂:0.065、0.195、0.780、2.340g·kg-1·d-1)考察甘遂对实验动物脏器系数、组织病理学、生化指标的影响,以及甘遂与甘草按不同比例配伍后毒性变化特点与规律,并通过因子分析方法综合分析甘遂-甘草反药组合的量-毒关系。结果:甘遂在《中华人民共和国药典》(以下简称药典)剂量范围内单独给药没有见到明显毒性,在药典高限4倍剂量下可产生肝、肾脏毒性;当甘遂在药典剂量范围内时,甘遂-甘草2∶1配伍没有见到明显毒性,1∶1、1∶2、1∶4、1∶10配伍可见不同程度的肝、肾、心脏毒性;当甘遂剂量超过药典规定剂量时,不同比例甘遂-甘草合用均增毒。通过因子分析从9个指标中抽提出3个公因子,共解释了78.0%的信息。甘遂-甘草配伍毒性可能随甘草比例的增加而增毒。结论:甘遂在其药典规定剂量范围内单用没有明显毒性,与甘草合用可表现出毒性,并随甘草比例的增加有毒性增加的趋势,说明在一定量比范围内,甘遂与甘草配伍相反。Objective: To investigate the dosage-toxicity relationship of the co-use of kansui and licorice, in order to explore the objective authenticity for their incompatibility and the regularity of the toxicity change. Methods: ICR mice were randomly divided into 27 groups: normal control group, kansui groups with different dosage, licorice groups with different dosage, and co-use groups with different ratio of kansui and licorice. Each group was adopted simultaneous to investigate the characteristic of toxicity within a high dosage range (0,065, 0.195, 0.780, 2.340g · kg^-1· d^-1 for kansui). The factor analytic approach was used to analyze the dosage-toxicity relationship of the co-use of kansui and licorice. Results: There was no obvious toxicity of kansui within the dosage range of pharmacopeia, while it had damages in liver and kidney with the dosage increased by 4 times. When it used within the dosage range of pharmacopeia, there was no obvious toxicity when the ratio of kansui and licorice was 2:1, but it had damages in liver, kidney and heart when the ratio of kansui and licorice were 1:1,1:2,1:4,1:10. The toxicity can be increased by the co-use of kansui and licorice. The factor analytic approach was used to extract three common factors from nine indexes. The results of the factor analysis suggested that the toxicity can be aggravated with the increased proportion of licorice. Conclusion: There is no obvious toxicity of the independent use of kansui within the dosage range of pharmacopeia. The toxicity can be aggravated with the increased proportion of licorice.
