摘 要:目的:观察两种补肾法对去势大鼠骨代谢和Ⅰ型胶原合成、交联及基质矿化相关基因表达的影响。方法:建立SD大鼠去势模型,给予补肾阳法和补肾阴法干预,观察骨组织形态学,检测骨密度及血清骨组织特异性碱性磷酸酶(B-ALP)、Ⅰ型前胶原氨基末端前肽(PINP)、Ⅰ型胶原氨基末端肽(NTX)和Ⅰ型胶原羧基末端肽(CTX),测定Ⅰ型胶原合成、交联与基质矿化相关调控基因COLIA-1、COLIA-2、Lhx9、Bglap、Ibsp、Runx-2和Smad3的表达。结果:补肾阳法和补肾阴法均可明显提高大鼠骨密度并改善骨组织微观结构,同时通过不同的方式影响骨代谢和Ⅰ型胶原合成、交联及基质矿化相关基因的表达。结论:补肾阴法和补肾阳法均可有效治疗大鼠绝经后骨质疏松症,但二者的作用机制存在明显差异。Objective: To observe the effects of two types of tonifying kidney on bone metabolism and gene expressions related to type I collagen synthesis, crosslink and matrix mineralization in ovariectomized (OVX) rats. Methods: OVX model of SD rats was established, and were intervened by tonifying kidney yang and tonifying kidney yin, Then the bone mineral density (BMD) was detected, the bone morphology was observed, serum levels of bone alkaline phosphotase (B-ALP), procollagen type I amino-terminal propeptide (PINP), N-telopeptide of type I collagen (NTX), C-telopeptide of type I collagen (CTX) were detected, and the expressions of type I collagen synthesis, crosslink and matrix mineralization related gene COLIA-1, COLIA-2, Lhx9, Bglap, Ibsp, Runx-2 and Smad3 were detected. Results: Both tonifying kidney yang and tonifying kidney yin could significantly increase BMD, ameliorate destruction of bone microstructure, affect bone metabolism, and change the expressions of type I collagen synthesis, crosslink and matrix mineralization related gene in different mechanisms. Conclusion: Both tonifying kidney yang and tonifying kidney yin are efficacy in treating rat postmenopausal osteoporosis, but the mechanism of two treatments are different.
