摘 要:目的:探讨补肾益智提取物对阿尔茨海默病(AD)模型小鼠学习记忆的影响及氧化应激、凋亡相关机制的调控。方法:采用腹腔注射东莨菪碱诱导的急性记忆障碍小鼠作为AD模型,补肾益智提取物(低、中、高剂量)和盐酸多奈哌齐进行干预。Morris水迷宫检测小鼠学习记忆能力,比色法检测血清氧化应激标志物超氧化物歧化酶(SOD)活力、丙二醛(MDA)和铜蓝蛋白(CP)含量,免疫组化法检测脑组织抗凋亡蛋白Bcl-x L表达,HE和Nissl染色观察脑组织病理形态变化。结果:与模型组相比,补肾益智方增加小鼠水迷宫平台穿越次数(P<0.01);提高血清SOD活力(P<0.05,P<0.01),降低血清MDA含量(P<0.05,P<0.01);增加Bcl-x L在海马和皮质区的表达(P<0.05,P<0.01),并改善脑组织的病理变化。结论:以补肾为靶点的益智提取物对东莨菪碱急性AD模型小鼠具有改善学习记忆能力的作用,其机制可能与抗氧化、抗凋亡的神经保护作用有关。Objective: To investigate the effects of Bushen Yizhi Formula extracts (BSYZ) on regulating learning and memory ability, oxidative stress and neuronal apoptosis in Alzheimer's disease (AD) mice. Methods: The model of AD was established with Intraperitoneal Injection of Scopolamine, mice were randomly divided into six groups: sham group, model group, BSYZ groups (low, medium and high doses) and Donepezil group. The animals' learning and memory ability were evaluated with Morris water maze test. The oxidant stress biomarkers superoxide dismutase (SOD), malondialdehyde (MDA) and ceruloplasmin (CP) in serum were detected by colorimetric method. The expression of anti-apoptotic protein Bcl-xL in brain was detected by immunohistochemistry. Haematoxylin- Eosin (HE) and Nissl staining were carded out to observe the histological changes in brain. Results: Compared with model group, BSYZ significantly increased the times of crossing the platform in space exploration test (P〈0.01). The serum SOD activity was improved (P〈0.05, P〈0.01), while the MDA level was decreased (P〈0.05, P〈0.01). Moreover, BSYZ up-regulated Bcl-xL expression in brain (P〈0.05, P〈0.01), and reversed the histological changes. Conclusion: BSYZ improves the memory impairment in Scopolamine-induced mice. This effect may be related to ameliorating oxidative stress and neuronal apoptosis.
