芍甘附子汤加味对Ⅱ型胶原诱导性关节炎大鼠OPG-RANKL表达的影响

摘　　要：目的:探讨芍甘附子汤(SGFZ)加味对CIA大鼠模型OPG、RANKL表达的影响。方法:选用牛Ⅱ型胶原与完全弗氏佐剂充分乳化后,于模型组大鼠左足跖及尾根部进行皮内免疫,并于第7天加强免疫,建立Ⅱ型胶原诱导型关节炎(CIA)模型。造模成功后,正常组和模型组给予0.9%氯化钠溶液灌胃,雷公藤组给予雷公藤多甙灌胃,芍甘附子汤加味组分别按小、中、大剂量灌胃干预;同时观察各组大鼠关节炎指数AI评分及足跖肿胀度,治疗结束后,观察其病理改变,并用Elisa法检测血清OPG、RANKL表达水平。结果:SGFZ大、中剂量组可减轻CIA大鼠关节肿胀度(P<0.01),减轻关节病理损害,上调OPG表达,降低RANKL、RANKL/OPG表达水平(P<0.01)。结论:芍甘附子汤加味对CIA大鼠关节骨破坏有保护作用,其机制可能通过调节OPG与RANKL表达水平实现的。Objective： To investigate the influence of modified Shaogan Fuzi Decoction （SGFZ） on expressions of OPG and RANKL of rats with collagen-induced arthritis （CIA）. Methods： Type Ⅱ collagen of beef was emulsified with complete Freund＇s adjuvant and used for intradermal immunization in left foot plantar and tail base of rats in model group, and enhanced immune on the 7th day, in order to established the CIA model. After animal models were established, rats in model group and control group received gavage administration with normal saline （0.9%NS）, rats in tripterygium wilfordii group received gavage administration with tripterygium wilfordii, and rats in SGFZ group received gavage administration with high, middle and low dosage of SGFZ. Arthritis index （AI） and toe swelling degree of rats in each group, and pathologic change after treatment were observed. The expression levels of osteoprotegerin （OPG） and receptor activator of NF-xB ligand （RANKL） were determined by using Elisa. Results： High and middle dosage of SGFZ decoction could relieve the toe swelling degree （P〈0.01） and the pathological lesion, up-regulate the OPG expression and down-regulate the expression of RANKL, RANKL/OPG （P〈0.01）. Conclusion： AGFZ could protect joint destruction, and the mechanism might be related with the regulation on the expression level of OPG and RANKL.
