摘  要:目的:研究以益气健脾为基础不同中医治法方药干预对实验性肿瘤恶病质小鼠肌肉萎缩的作用及其可能的机制。方法:实验分为健脾和胃组,益气温阳组,健脾补血组,健脾解毒组,益气养阴组,正常组1、2,及荷瘤组1、2;BALB/c小鼠皮下种植CT26细胞后,治疗15d,分别对各组心肌、腓肠肌、附睾脂肪及瘤体称重比较质量,HE染色检测其肌肉形态,蛋白免疫印迹法(Western blotting)检测肌环指蛋白1(MURF1)表达,免疫组化检测对肌肉萎缩抑制明显的治疗组肌肉组织核转录因子(NF-κBp65)表达及磷酸化核转录因子(phospho-p65)水平。结果:荷瘤组各脏器质量下降明显,腓肠肌组织肌纤维降解,MURF1、NF-κBp65蛋白表达上调及phospho-p65水平增加;与荷瘤组1比较,健脾解毒组腓肠肌、心肌及附睾脂肪质量减少明显(P<0.05),益气温阳组心肌重量减少(P<0.05),健脾补血组腓肠肌及附睾脂肪重量减少(P<0.05),健脾解毒组及健脾补血组MURF1表达下调,健脾解毒组接近正常;同时,健脾解毒组NF-κBp65蛋白表达及phospho-p65蛋白水平下调,肌纤维形态接近正常。结论:以益气健脾为基础的不同中医治法中,健脾解毒方药能够有效抑制实验性肿瘤恶病质肌肉消耗,抑制疾病进展过程中的炎性反应是其可能的机制之一。Objective: To study effects and probable mechanisms of formulas based on different principles of traditional Chinese medicine on cachexia muscle atrophy of mice induced by CT26 adenocarcinoma. Methods: Invigorating spleen for harmonizing stomach group, warming yang and invigorating spleen group, invigorating spleen and replenishing blood group, invigorating spleen and removing toxicity group, benefiting qi and nourishing yin group, control group 1 and 2, and model group 1 and 2 were set up in this research. CT26 cells were hypodermic implanted in BALB/C mice, and these mice were treated with corresponding treatment for 15 days. The weight of heart, gastrocnemius muscle, epididymisadipose mass and tumor of mice in each group were weighed and compared. The muscle morphological was observed by using HE staining, and the expression of MURF1 was test by using western blot. Immunohistochemical was used to test expression of NF-xBp65 and phospho-p65. Results: The weights of organs of mice in model group were decreased significantly. The muscle fibers of gastrocnemius muscle were dissolved, and the expression of MURF1 and NF-xBp65 was up-regulated, while dephosphorylation of NF-κBp65 was increased of mice in model group. Compared with model group I, the weights of gastrocnemius muscle, heart and epididymisadipose massof mice in invigorating spleen and removing toxicity group were reduced significantly (P〈0.05), and the weights of heart of mice in warming yang and invigorating spleen group and weights of epididymisadipose mass and gastrocnemius muscle of mice in invigorating spleen and replenishing blood group were also reduced significantly (P〈0.05). The expression of MURF1 of mice in invigorating spleen and replenishing blood group and invigorating spleen and removing toxicity group was down-regulated, and those of mice in invigorating spleen and removing toxicity group near to that of control group.
