摘 要:目的:研究黄芩水提液干预对呼吸道合胞病毒(RSV)感染BALB/c小鼠血浆和肺组织脂质代谢异常的调节作用。方法:RSV滴鼻感染BALB/c小鼠,黄芩水提液灌胃给药进行干预,采集血浆和肺组织样品进行基于超高效液相-二维线性离子阱高分辨静电场组合质谱仪(UPLC-LTQ/Orbitrap XL)的脂质组学研究,考察干预前后脂质代谢物组的变化,寻找潜在生物标志物,探讨黄芩水提液抗病毒作用的途径。结果:正常组、模型组、利巴韦林组、黄芩组血浆和肺组织脂质谱均得到明显分离,在血浆和肺组织中分别鉴定了与RSV感染相关的9种和7种生物标志物,主要包括甘油磷脂酰胆碱、鞘磷脂和甘油三酯,黄芩水提液干预对部分异常脂质代谢具有明显的改善作用。结论:黄芩水提液能有效缓解RSV感染所致的甘油磷脂酰胆碱、鞘磷脂和甘油三酯代谢紊乱,表明黄芩抗病毒的作用机制可能与调节这些代谢途径有关。Objective: To study the regulation effects of baical skullcap root extraction on abnormal lipids metabolism in plasma and lung of mice infected with respiratory syncytial virus (RSV). Methods: Mice received intranasal administration with RSV and gavage administration with baical skullcap root extraction. Blood and lung tissue were collected and used to test the lipidomics by using UPLC-LTQ/Orbitrap XL, in order to observe the changes of lipid metabolites before and after intervention, look for the potential biomarkers and discuss the functional channel of balcal skullcap root extraction. Results: The blood and lung tissue of mice in control group, model group, ribavirin group and baical skullcap root group were separated evidently. There were 9 biomarkers associated with RSV in blood and 7 biomarkers associated with RSV in lung tissue, which mainly included acetyl glyceryl ether phosphatidylcholine (AGEPC), sphingomyelin and triglyceride. Intervention of baical skullcap root extraction could improve part of abnormal lipid metabolism significantly. Conclusion: Baical skullcap root extraction could relieve metabolic disorder of AGEPC, sphingomyelin and triglyceride effectively, and it shows that the anti-virus mechanism of baical skullcap root extraction might be related with regulation on these metabolic pathways.
