摘 要:目的:探讨中药黄芩乙醇提取物对血管性痴呆(Va D)沙土鼠的保护作用及抑制海马CA1区神经细胞凋亡的研究。方法:以双侧颈总动脉夹闭沙土鼠为研究对象,实验分为正常对照组、Va D组、Va D+黄芩低剂量(25mg/kg)组、Va D+黄芩中剂量(50mg/kg)组、Va D+黄芩高剂量(100mg/kg)组,采用乙醇提取黄芩,腹腔注射给药,2次/d,共14d。采用Morris水迷宫评价沙土鼠的学习记忆能力,Nissl染色观察海马CA1区神经细胞的形态变化,TUNEL染色观察神经细胞的凋亡,Western blotting方法检测B细胞淋巴瘤-2(Bcl-2)/Bcl-2相关x蛋白(Bax)凋亡通路变化。结果:黄芩乙醇提取物能够增加提高沙土鼠的学习记忆能力,减少Morris水迷宫中潜伏期时间(P<0.05,P<0.01),增加穿梭次数(P<0.05,P<0.01);减轻海马组织的病理性改变;降低神经细胞的凋亡数(P<0.01),并且提高了Bcl-2/Bax的比值(P<0.05)。结论:黄芩乙醇提取物能够改善Va D沙土鼠的学习记忆能力,减轻海马组织的凋亡,其作用机制可能与Bcl-2/Bax凋亡通路相关。Objective: To investigate the protective effective of ethanol extract of baical skullcap root on gerbils with vascular dementia(VaD) and inhibition effect of nerve cell apoptosis in hippocampal CA1 area. Methods: Gerbils with bilateral carotid artery clipping were as the object of study. Control group, model group, low dosage of baical skullcap root group (25mg/ kg), middle dosage of baical skullcap root group (50mg/kg), and high dosage of baical skullcap root group (100mg/kg). Baical skullcap root was extracted by using ethyl alcohol, and gerbils were treated with inta'aperitoneal injection 2 times a day and lasted for 14 days. Learning and memory ability were evaluated by using Morris water maze. Nissl staining was used to observe the morphological changes of nerve ceils in hippocampal CA1 area. TUNEL staining was used to observe nerve cell apoptosis, and Western blotting was used to test the changes of Bcl-2/Bax apoptosis pathway. Results: Ethanol extract of baical skullcap root could improve the ability of learning and memory of gerbils, reduce the latency time in Morris water maze (P〈0.05, P〈0.01), increase travel times (P〈0.05, P〈0.01), reduce the pathological changes of hippocampal tissue, decrease the apoptosis number of nerve cells (P〈0.01), and increase the ration of Bcl-2/Bax (P〈0.05). Conclusion: Ethanol extract of baical skullcap root couldimprove the ability of learning and memory of VaD gerbils and alleviate apoptosis of hippocampal tissue, and the mechanism might be related with apoptosis pathway of Bcl-2/Bax.
