摘 要:为有效提高宗地花猪的肉品质,合理评价养殖模式提供理论依据,参照GenBank中猪H-FABP基因序列设计引物,以猪18SRNA为内参基因,应用实时荧光定量PCR方法检测H-FABP基因在不同饲养条件下宗地花猪不同组织器官中的表达量。结果表明:H-FABP基因在肺脏、心脏、肾脏、小肠、脾脏、肝脏、背最长肌、腰大肌中均有表达,但不同饲养条件下其表达水平各异,放养型背最长肌>腰大肌>肾脏>心脏>小肠>肺脏>脾脏>肝脏,背最长肌中表达量显著高于其他组织(P<0.05);圈养型腰大肌>脾脏>心脏>肺脏>小肠>肝脏>背最长肌>肾脏,腰大肌中表达量显著高于其他组织(P<0.05),肝脏中表达量高于背最长肌(P>0.05)。两种饲养条件下宗地花猪肺脏、肝脏中表达量相当,而在心脏、肾脏、小肠、脾脏、背最长肌中表达差异较大。The expression quantity of H-FABP gene in different organs of Zongdi spotted pig under different feeding patterns was detected by real-time fluorescence quantification PCR method with specific designed primers according to swine H-FABP gene in GenBank and by using swine 18S RNA as the reference gene to provide the theoretical basis for effectively improving meat quality of Zongdi spotted pig and rationally evaluating breeding pattern.The results showed that H-FABP gene has expression in lung, heart,kidney,small intestine,spleen,liver,longissimus muscle and psoas,but there is difference in the expression level between two different breeding patterns.The H-FABP expression quantity in different organs of Zongdi spotted pig under the stocking pattern is longissimus muscle〉psoas〉kidney〉heart〉small intestine〉lung〉spleen〉liver and the H-FABP expression quantity in longissimus muscle is higher than other organs significantly(P〉0.05).The H-FABP expression quantity in different organs of Zongdi spotted pig under the captive pattern is psoas〉 spleen〉heart〉lung〉 small intestine〉 liver〉longissimus muscle〉kidney.The H-FABP expression quantity in psoas is higher than other organs significantly(P〈0.05)and the H-FABP expression quantity in liver is higher than longissimus muscle (P 〉0.05).There is no obvious difference in H-FABP expression quantity of lung and liver between two feeding patterns but difference in H-FABP expression quantity of heart,kidney,small intestine,spleen and longissimus muscle between two feeding patterns.
