摘 要:该文应用1.5,2.0,3.0 g/L亚硫酸钠制造新的肺动脉平滑肌细胞化学缺氧模型,并与三气培养箱制造的物理缺氧模型进行对比,采用hematoxylin-eosin staining(HE染色)、Cell Counting Kit-8(CCK-8)来观察亚硫酸钠对细胞的影响,检测细胞体内的活性氧和缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)的基因及蛋白表达量来判断模型的缺氧程度。细胞体内活性氧和HIF-1α的检测结果提示,亚硫酸钠所制造的缺氧模型的缺氧程度随其浓度的升高而升高,虽然2.0 g/L和3.0 g/L亚硫酸钠组对细胞有一定程度的毒性作用,但是1.5 g/L亚硫酸钠组对细胞的损害程度与物理缺氧组相比无明显差异。该研究表明,1.5 g/L亚硫酸钠所制造的肺动脉平滑肌细胞化学缺氧模型较符合肺动脉平滑肌细胞缺氧模型的要求。Different concentrations of sodium sulfite of 1.5, 2.0, 3.0 g/L were used to make a new chemical hypoxia model of pulmonary artery smooth muscle cells(PASMCs). Hematoxylin-eosin(HE) staining and Cell Counting Kit-8(CCK-8) were performed to observe the effect of sodium sulfite on the PASMCs. To define the anoxic extent of new hypoxia model, the changes of reactive oxygen species and the expression of HIF-1α were detected. The content of ROS tested by fluorescence microscope and the expressions of HIF-1α tested by RT-PCR and Western blot indicated that the levels of hypoxia were dependent on the concentration of the sodium sulfite. Although there were some damages to the PASMCs in the hypoxia model induced by 2.0 g/L and 3.0 g/L, but the model induced by 1.5 g/L of sodium sulfite was similar to the model induced by three gas incubator. So the concentration of sodium sulfite with 1.5 g/L was the best to induce a hypoxia model.
