摘 要:目的探讨孕期边缘型维生素A缺乏(MVAD)对新生鼠海马中视黄酸核受体α(RARα)、肉瘤基因Src和N-甲基-D-天门冬氨酸受体1(NMDAR1)表达的影响。方法构建孕期维生素A正常(VAN)和MVAD大鼠模型(VAN组和MVAD组);分离培养新生鼠原代海马神经元,免疫荧光染色鉴定;采用real-time PCR和Western blotting检测新生鼠海马组织及体外培养的原代海马神经元中RARα、Src和NMDAR1的mRNA和蛋白表达;利用钙影像仪检测原代海马神经元钙兴奋性。结果免疫荧光染色显示,分离培养的新生鼠原代海马神经元95%表达神经元标志物神经元特异性烯醇化酶(NSE);MVAD组新生鼠海马组织和原代海马神经元中RARα、Src、NMDAR1的mRNA和蛋白表达水平明显低于VAN组(P<0.05);MVAD组新生鼠海马神经元的钙兴奋性明显低于VAN组(P<0.05)。结论孕期MVAD会降低海马RARα、Src和NMDAR1的表达,可能与孕期MVAD影响幼鼠的学习记忆功能有关。Objective To explore the marginal vitamin A deficiency( M V AD) in pregnancy on the expressions of retinoic acid receptor alpha( RARα), sarcoma gene Src, and N-methyl-D-aspartate receptor subunit 1( N M DAR1) in the hippocampus of newborn rats.Methods The pregnant rat models of vitamin A normal( V AN group) and marginal vitamin A deficiency( M V ADgroup) were established.The hippocampus tissue of newborn rats was isolated.The primary hippocampus neurons were cultured in vitro and detected by the immunofluorescence staining.The mRN A and protein expressions of RARα, Src, and N M DAR1 in hippocampus tissue and primary hippocampus neurons of newborn rats were detected by the real-time PCR and W estern blotting.The calciumexcitability of primary hippocampus neurons was detected by the calciumimaging instrument.Results The results of immunofluorescence staining showed that 95% of primary hippocampus neurons of newborn rats that were isolated and cultured in vitro expressed the neuron maker of neuron-specific enolase( N SE).The mRN A and protein expressions of RARα, Src, and N M DAR1 in hippocampus tissue and primary hippocampus neurons of newborn rats of the M V ADgroup were significantly lower than those of the V AN group( P〈0.05).The calciumexcitability of hippocampus neurons of newborn rats of the M V ADgroup was significantly lower than that of the V AN group( P〈0.05).Conclusion M V ADin pregnancy can decrease expressions of RARα, Src, and N M DAR1 in the hippocampus.This may be relevant to the impaired learning and memory functions of newborn rats caused by M V ADin pregnancy.
