胎儿脐带血淋巴细胞亚群表达特性

摘　　要：目的确定正常胎儿脐带血淋巴细胞亚群的百分比,并与正常儿童外周血淋巴细胞亚群进行比较,从而探析胎儿脐带血淋巴细胞亚群表达特性。方法收集怀孕25^(+1)周~26^(+3)周胎儿的脐带血12例,排除染色体异常等遗传性疾病后,采用流式细胞仪检测其淋巴细胞表面标记CD3^+、CD9^+、CD16^+CD56^+、CD4^+、CD8^+、CD25^+、CD38^+、CD3^+HLA-DR^+、CD8^+HLA-DR^+、CD8^+CD38^+和CD4^+CD25^+表达比例,同时检测22例健康儿童外周血的淋巴细胞表面标记,对结果进行比较。结果与正常儿童外周血比较,胎儿脐带血淋巴细胞CD3^+、CD4^+、CD4^+CD25^+、CD3^+CD25^+、CD38^+表达显著升高,而CD19^+、CD16^+CD56^+、CD8^+HLA-DR^+、CD3^+HLA-DR^+、CD8^+HLA-DR^+表达则下降(P<0.01);而CD8+CD38+淋巴细胞表达相差不大。结论本研究确定了正常胎儿脐带血中淋巴细胞的表达情况,表明胎儿脐带血均处于幼稚阶段,T细胞升高,B细胞、NK细胞相对降低,而一些活化的T细胞减少,调节性T细胞升高。Objective To explore the percentage of lymphocyte subsets in normal fetal umbilical cord blood, and compare it with that of normal peripheral blood lymphocyte subsets in children, so as to study fetal umbilical cord blood lymphocyte subsets expression characteristics. Methods 12 samples of fetal umbilical cord blood were collected with the pregnancy from 25 ＋1 weeks to 26 ＋3 weeks. Structural chromosomal abnormalities were eliminated. FACS was used to detect the proportions of CD3 ^＋ , CD19^＋ , CD16^＋CD56^＋, CD4^＋ , CD^＋ , CD25^＋ , CD38^＋ , CD3^＋HLA-DR^＋ , CD8 ^＋ HLA-DR^＋ , CD8^＋CD38^＋ and CD4^＋CD25 ^＋ cells. 22 samples of peripheral blood of healthy children were also detected as normal control. Results Compared with normal control, the proportions of CD3 ^＋ , CD4^＋ , CD4 ^＋ CD25 ^＋ , CD3 ^＋CD25 ^＋ , CD38^＋ in fetal umbilical cord blood lympho- cytes significantly increased, while the expression of CD19, CD16 ^＋ CD56 ^＋, CD8 ^＋ , HLA - DR ^＋ , CD3 ^＋ HLA - DR ^＋, CD8^＋ HLA- DR ^＋ declined（P 〈 0.01 ）. No significant difference was found in CD8^＋ CD38 ^＋ cells. Conclusion This study deter- mined the expression of lymphocytes of normal fetal umbilical cord blood of fetal umbilical cord blood, which shows that it is in the immature stage, T cells increased, B cells and NK cells were relatively lower, and some activated T ceils reduced, regulato- ry T cells increased.
