摘 要:目的研究神经生长因子(NGF)在支气管哮喘气道重塑中的作用及其可能机制,为哮喘治疗寻求新的靶点。方法将30只SPF级健康雄性Wistar大鼠随机分为对照组、哮喘模型组和NGF抗体干预组。苏木精-伊红染色观察大鼠气道壁组织学改变,Masson染色观察气道上皮下胶原层厚度,免疫组化法检测肺组织血管内皮生长因子(VEGF)、Ⅲ型胶原表达。结果哮喘模型组及NGF抗体干预组支气管壁厚度、支气管平滑肌厚度、支气管平滑肌细胞核数、支气管壁血管密度、上皮下胶原层厚度、VEGF及Ⅲ型胶原的表达较对照组显著增加,NGF抗体干预组以上改变较哮喘模型组减轻,差异均有统计学意义(F=108.29~554.21,P〈0.01)。大鼠支气管壁血管密度和管壁厚度与肺组织中VEGF蛋白表达,气道胶原层厚度与Ⅲ型胶原含量均呈正相关(r=0.82~0.87,P〈0.05)。结论 NGF可能通过调控肺组织VEGF、Ⅲ型胶原的表达参与哮喘气道重塑。Objective To study the effect of nerve growth factor(NGF)on the airway remodeling of asthma and its possible mechanisms,and explore new targets for its therapy. Methods Thirty healthy male Wistar rats were divided into normal group,the asthma model group and NGF-antibody group at random.HE staining was used to observe the histological changes of the airway,Masson staining to estimate the thickness of epithelium collagen,and immunohistochemistry to measure the levels of VEGF and collagen Ⅲ. Results The thickness of the bronchial wall and the bronchial smooth muscle(BSM),the number of the BSM cells,the vascular density of the bronchial wall,the thickness of epithelium collagen,the levels of VEGF and collagenⅢ in the asthma model group and the NGF-antibody group increased versus the control group,and the above changes in the NGF-antibody group were lighter than the asthma model group,the differences being significant(F=108.29-554.21,P0.01).The density of the bronchial vessels and the thickness of the wall were positively correlated with VEGF expression in the lung tissue,the thickness of the bronchial wall and the level of collagen Ⅲ(r=0.82-0.87,P0.05). Conclusion Nerve growth factor is likely involved in airway remodeling of asthma rats by regulating the expressions of VEGF and collagen Ⅲin lung tissue.
