SV40启动子在烟草体内的驱动特性研究

摘　　要：根据SV40启动子序列设计引物,PCR扩增该启动子后,成功构建了具有SV40启动子和GUS报告基因的植物表达载体pLpPSG,并将重组质粒转化到烟草叶片中.通过SV40启动子驱动的GUS基因在烟草叶片内的瞬时性表达,研究了该启动子在植物基因表达中的驱动特性.结果表明:SV40启动子可启动GUS基因在烟草体内的表达,其提高基因表达水平达到2×35S启动子的(88.5±19.2)%.Based on the SV40 promoter sequence,apair of primers was designed and SV40 promoter was cloned by PCR.A recombinant expression plasmid pLpPSG,which contains SV40 promoter and a GUS reporter gene,was constructed.And then the constructed expression vector was transformed into tobacco leaves.The promoting characteristics of the SV40 promoter in tabaco were investigated by the transient expression of GUSgene driven by SV40 promoter in tobacco leaves.The results showed that SV40 promoter promoted the GUS expression in tobacco.The expression level of the GUSreporter gene driven by SV40 promoter achieved（88.5±19.2）% of the expression level of GUSgene driven by 2×35Spromoter.
