Effects of hydrogen-rich saline on Akt/GSK3β signaling pathways and cardiac function during myocardial ischemia-reperfusion in rats

摘  要:目的 探讨饱和氢盐水对心肌缺血再灌注损伤大鼠心肌细胞Akt/GSK3β信号通路和心功能的影响.方法 选取健康成年清洁级雄性SD大鼠150只,采用随机数字表法将其分为5组(n=30):空白对照组(Ⅰ组)、假手术组(Ⅱ组)、缺血再灌注组(Ⅲ组)、饱和氢盐水治疗组(Ⅳ组)、生理盐水治疗组(Ⅴ组).Ⅰ组不做任何处理;Ⅱ组大鼠仅穿线不结扎;Ⅲ组制备大鼠心肌缺血再灌注损伤模型;Ⅳ组于再灌注前5 min给予1 ml/100 g腹腔注射饱和氢生理盐水;Ⅴ组于再灌注前5 min给予1 ml/100 g腹腔注射等量生理盐水.采用BL-420型生物机能实验系统记录大鼠缺血前、缺血30min、再灌后60、120 min,4个时间点的正负左室内压最大变化速率(±dp/dt max),左室舒张末期压(LVDP)及左室收缩末期压(LVSP)数据.于再灌注60、120 min后取左室心肌组织HE染色观察心肌病理学变化.采用蛋白质印迹法检测心肌细胞蛋白激酶B(Akt)和糖原合成酶激酶-β(GSK3β)的表达.结果 与Ⅰ组、Ⅱ组比较,Ⅲ组、Ⅳ组和Ⅴ组大鼠缺血30 min,再灌注60、120 min时+dp/dt max(F=24.61,26.55,25.21),-dp/dt max(F=20.27,18.57,19.35) LVDP(F=23.53,21.79和19.47)及LVSP(F =21.72,17.75和18.22)显著降低,心肌细胞Akt(F=18.62,17.41)和GSK3β(F=15.27,15.58)表达水平升高.与Ⅲ组、Ⅴ组比较,Ⅳ组大鼠+dp/dt max(F=24.52和26.95),-dp/dt max(F =22.71和23.18),LVDP(F =28.56和26.42)及LVSP(F=23.18和25.39)显著升高,心肌细胞Akt(F=17.89和17.43)和GSK3β(F=14.85和14.29)表达水平降低.与T0比较,Ⅲ组、Ⅳ组和Ⅴ组T1-3+ dp/dt max(F=20.15,22.49和23.12),-dp/dt max(F =20.37,21.83和23.59),LVDP(F=22.38,21.86,22.75)及LVSP(F=25.01,24.21和25.76)显著下降.与T1比较,Ⅳ组T2-3+ dp/dt max(F =27.15),-dp/dt max(F =22.87),LVDP(F=16.81)及LVSP(F=26.14)显著上升.与T2比较,Ⅳ组T3心肌细胞Akt(F=14.97)和GObjective To explore the effects of hydrogen-rich saline on Akt/GSK3β signaling pathways and cardiac function during myocardial ischemia-reperfusion (I/R) in rats.Methods A total of 150 healthy male Sprague-Dawley rats were selected and divided randomly into 5 groups of normal control (Ⅰ),sham operation (Ⅱ),myocardial ischemia-reperfusion (Ⅲ),hydrogen-rich saline (Ⅳ) and normal saline group (Ⅴ) (n =30 each).Group Ⅰ had no treatment at all.In group Ⅱ,anterior descending branch was merely exposed but not ligated.Myocardial I/R was induced by 30-min occlusion of anterior descending branch of left coronary artery followed by 12/24 h reperfusion of Bimbaum.Hydrogen-rich saline 1 ml/100 g were injected intraperitoneally 5 min before reperfusion in group Ⅳ.Normal saline 1 ml/100 g was injected intraperitoneally 5 min before reperfusion in group Ⅴ.The values of ± dp/dt max,left ventricular diastolic pressure (LVDP) and left ventricular systolic pressure (LVSP) during the procedure were recorded by BL-420 biological function experimental system at pre-ischemia,ischemia 30 min,60 min and 120 min of reperfusion.The animals were sacrificed at 60 min,120 min of reperfusion and hearts harvested.The pathological changes of myocardial tissue were detected by hematoxylin and eosin staining.And the expressions of Akt and GSK3β in myocardial tissue were examined by Western blot.Results Compared with groups Ⅰ and Ⅱ,the values of ± dp/dt max,LVDP and LVSP significantly increased.And the expressions of Akt and GSK3β in myocardial tissue significantly increased in groups Ⅲ,Ⅳ and Ⅴ.Compared with groups Ⅲ and Ⅴ,the values of ± dp/dt max,LVDP and LVSP significantly decreased.And the expressions of Akt and GSK3β in myocardial tissue significantly decreased in group Ⅳ.Compared with T0,the values of ± dp/dt max,LVDP and LVSP significantly decreased at T1-3 in groups Ⅲ,Ⅳ and Ⅴ.Compared with T1,the values of ± dp/dt max,LVDP and LVSP significantly increased at
