摘 要:【目的】通过银屑平颗粒对豚鼠银屑病模型全血粘度、干扰素-γ(Interferon-γ,IFN-γ)、白细胞介素-10(interleukin-10,IL-10)的影响,进一步探讨银屑平颗粒治疗银屑病的可能作用机理。【方法】通过采用ELISA法测定豚鼠耳部皮损组织匀浆中细胞因子IFN-γ、IL-10含量,右耳用作病理组织学检查,各组病变程度以秩和检验进行组间比较。观察银屑平颗粒对心得安诱导的豚鼠银屑病模型全血粘度及IFN-γ、IL-10的影响。【结果】心得安诱导的豚鼠银屑病模型在高切、中切、低切变率下(180.0、100.0、1.0/S)全血粘度(3.15±0.37,3.40±0.51,12.00±4.75)较正常对照组(2.58±0.19,2.62±0.25,6.44±1.81)显著增高,各给药组均可降低各切变率下的全血粘度,其中银屑平颗粒高(2.45±0.21,2.46±0.16,7.71±1.80)、中(2.64±0.19,2.75±0.32,7.63±2.09)、低剂量(2.65±0.21,0.75±0.32,6.53±1.50)和阳性对照药银屑灵组(2.80±0.20,2.90±0.33,8.06±1.99)与模型组(3.15±0.37,3.40±0.51,12.00±4.75)相比有显著性差异(P<0.05)。银屑平颗粒高(55.927±4.608,76.648±9.945)、中剂量组(56.642±4.450,75.035±9.699)以及阳性对照药甲氨喋呤(56.768±4.575,77.443±9.587)、银屑灵组(55.378±6.797,74.873±5.880)可明显地降低心得安诱导的豚鼠银屑病模型耳组织匀浆中IFN-γ含量,增加保护性细胞因子IL-10的含量,与模型组(62.917±6.646,62.917±6.646)相比差异有显著性(P<0.05)。【结论】银屑平颗粒可能通过改善血液流变学和调整机体的免疫异常发挥其治疗银屑病的功效。【Objective】On the basis of the previous researches, this paper is to explore the possible mechanism of Yinxieping granules,throughing investigating the effects of Yinxieping granules on blood viscosity and cytokine levels of psoriasis model in guinea pigs induced by propranolol hydrochloride.【Methods】The contents of IFN-γ and IL-10 in homogenate of the lesional skin of the ear were determined using the ELISA method. And the right ears were tested in the pathological and histological examination.The rank-sum test was used to compare the difference between groups.【Results】The results showed that compared with control group(2.58±0.19,2.62±0.25,6.44±1.81),blood viscosity of experimental groups improved significantly at high,medium and low shear rates(180.0、100.0、1.0/s)in cavia procellus psoriasis model(3.15±0.37,3.40±0.51,12.00±4.75). Compared with yinxueling(2.80±0.20,2.90±0.33,8.06±1.99),blood viscosity of yinxueping granules also improved significantly at high(2.45±0.21,2.46±0.16,7.71±1.80),medium(2.64±0.19,2.75 ± 0.32,7.63 ± 2.09) and low(2.65 ± 0.21,0.75 ± 0.32,6.53 ± 1.50) dosage(P〈0.05). High dosage of Yinxieping granules(55.927±4.608,76.648±9.945),middle dosage(56.642±4.450,75.035±9.699),positive control drug of methotrexate(56.768±4.575,77.443±9.587) and Yinxieling(55.378±6.797,74.873±5.880) can evidently decrease the content of IFN- γ in homogenate of the lesional skin of the ear of the psoriasis cavy models and increase the content of the protective cell factor of IL-10, significant differences can be observed when compared with the model group(3.15 ± 0.37,3.40 ±0.51,12.00 ± 4.75)(P〈0.05).【Conclusions】It indicated that Yinxieping granules may act on psoriasis through a possible way by ameiliorating the hemorrheology and adjusting the immune abnormalities of the organism.
