摘 要:目的 探讨清热活血颗粒剂对单纯熏烟法建立大鼠慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)模型气道重塑的影响.方法 采用单纯熏烟法造模,将SPF级54只健康雄性SD大鼠随机分为正常组、COPD模型组、布地奈德组、清热活血颗粒(低、中、高)剂量组,每组9只.给予相应处理36 w,观察HE染色后大鼠肺部病理变化;小动物肺功能检测系统评估实验大鼠肺功能;ELISA测定实验大鼠血清MMP-9、TIMP-1以及TGF-β水平;Western blot法检测肺组织MMP-9、TIMP-1及TGF-β的表达水平.结果 清热活血颗粒各剂量组均不同程度延缓肺功能下降,使FVC降低,使FEV0.1/FVC、MMEF及PEF升高,改善气道重塑的病理改变,优于布地奈德组(P<0.01).清热活血颗粒各剂量组均能一定程度降低大鼠血清细胞因子TGF-β水平,恢复MMP-9/TIMP-1平衡.结论 清热活血颗粒能够明显延缓COPD模型大鼠的肺功能下降,改善气道重塑.Objective To study the effect of early intervention with Qingre huoxue granules on airway remodeling in COPD rats with the method of establishing the smoke-mediated rat model of COPD. Methods Totally 54 healthy male Sprague Dawley (SD) rats were randomly divided into normal control group, model group, budesonide group, and the Qingre huoxue granules (low, middle and high) dose group with nine rats in each group. After 36 weeks treatment, The morphology changes of the lungs were observed with HE staining, and the pulmonary function was evaluated by BUXCO system. The serum MMP - 9 and TIMP - 1, TGF - beta level were detected using enzyme-linked immunosorbent assay; Arterial blood gas was detected by virtue of blood gas analyzer. Results Compared with Budesonide group, Qingre huoxue granules of each dose group can help better improve the forced vital capacity (FVC), FEV0.1 / FVC, maximum mid ex- piratory flow rate (MMEF), forced expiratory flow (PEF) (P〈0.01) as well as amelioratethe pathological damage of COPD. Compared with normal control group, the level of MMP - 9and TIMP - 1 were significantly increased (P〈0.05 or P〈0.01) in the serum of model group. Compared with model group, MMP - 9 and TIMP - 1 in the Qingre huoxue granules of each dose group were gradually reduced (P〈0.05 or P〈 0.01). Conclusions Qingre huoxue granules could significantly slowed down the speed of declining in lung function of rats, as well as improved the condition of airway remodeling.
